酸枣仁汤对抑郁模型大鼠海马DKK-1与β-catenin、GSK-3β的影响 被引量:6

摘要 目的:探讨酸枣仁汤的抗抑郁作用及机制。方法:48只大鼠按随机数字表法分为空白组、模型组、酸枣仁汤低、中、高剂量组及氟西汀组,采用慢性轻度不可预见性应激制备抑郁大鼠模型,酸枣仁汤与氟西汀连续治疗28 d后观察各组大鼠体质量、糖水消耗、旷场实验得分,免疫组化法检测海马中DKK-1蛋白表达,荧光定量PCR法检测海马组织中β-catenin及GSK-3β的mRNA表达。结果:与正常组比较,模型组大鼠体质量、糖水偏爱百分比和旷场试验得分均显著降低(P<0.01),海马区DKK-1蛋白表达明显上升(P<0.01),GSK-3β的mRNA表达明显下降(P<0.01);与模型组比较,酸枣仁汤中、高剂量组、氟西汀组较模型组体质量、糖水偏爱百分比和旷场试验得分均明显升高(P<0.05,P<0.01),酸枣仁汤低、中、高剂量组及氟西汀组大鼠海马中DKK-1蛋白表达显著下降(P<0.01),酸枣仁汤高剂量组大鼠海马中β-catenin、GSK-3βmRNA表达明显上升(P<0.05,P<0.01)。结论:酸枣仁汤能够调节抑郁模型大鼠海马组织中DKK-1蛋白与β-catenin、GSK-3βmRNA表达,其抗抑郁机制可能与对海马神经元的保护作用有关。 Objective:To explore the antidepressant effect and mechanism of Suanzaoren decoction.Methods:48 rats were divided into six groups:blank group,model group,Suanzaoren decoction low,medium and high dose groups and fluoxetine group according to the random number table method.Depression rat models were prepared by chronic mild unpredictable stress.Suanzaoren decoction and fluoxetine were continuously treated for 28 days.Observe the body weight,sugar consumption,and open field test scores.Immunohistochemistry was used to detect the expression of DKK-1 protein in the hippocampus.Rt-qPCR method was used to detect the mRNA expression ofβ-catenin and GSK-3βin hippocampus.Results:Compared with the blank group,the body mass,sugar water preference and open field test scores of the model group were significantly lower(P<0.01),and the expression of DKK-1 protein in hippocampus was significantly increased(P<0.01),GSK-3βmRNA expression was decreased significantly(P<0.01).Compared with the model group,the body mass,sugar water preference and open field test scores of the middle and high dose groups of Suanzaoren decoction and fluoxetine group were significantly higher(P<0.05,P<0.01),the expression of DKK-1 protein in the hippocampus of rats in the low,medium and high dose groups of Suanzaoren decoction and fluoxetine group was significantly lower(P<0.01),and the expression ofβ-catenin、GSK-3βmRNA in the hippocampus of rats in the high dose group of Suanzaoren decoction was significantly increased(P<0.05,P<0.01).Conclusion:Suanzaoren decoction can affect the expression of DKK-1 protein andβ-catenin,GSK-3βmRNA expression in the hippocampus of depression model rats,and its antidepressant mechanism may be related to the protective effect of hippocampal neurons.
