响应面法优化产木聚糖酶耐热菌株的发酵条件 被引量:3

Response Surface Optimization of Fermentation Conditions for Xylanase Production by a Thermotolerant Strain

摘要 通过选择性培养基分离到一株高产木聚糖酶耐热菌株XY1,研究了其所产木聚糖酶的最适反应温度、最适pH值以及发酵条件的优化。结果表明,木聚糖酶的最适反应温度为60℃、最适pH值为5.5,利用响应面中的BOX-Behnken优化产木聚糖酶的最适发酵条件为麸皮22.25 g/L、NH_(4)Cl 5.05 g/L、发酵时间26.16 h。在此发酵条件下,所产酶的酶活为139.37 U/mL,比未优化前提高了29.05%。 A heatresistant xylanase producing strain XY1 was isolated by selective medium.The effect of temperature and pH on the activity of crude xylanase from XY1 was studied.The results showed that the optimal temperature and pH for the mannanase activity were 60℃and 5.5,respectively.Box-Behnken design of response surface methodology was used to optimize the fermentation conditions of xylanase.The optimum fermentation conditions were wheat bran 22.25g/L,NH_(4)Cl 5.05 g/L and fermentation time 26.16 h.Under the optimal fermentation conditions,the maximum enzyme activity was 139.37 U/mL.The maximum enzyme activity increased with approximately 29.05%.
