摘要
目的比较浙产金线莲水提液和醇提液对急性酒精性肝损伤小鼠肝组织中白介素1β(IL-1β)、白介素6(IL-6)表达的影响,初步探讨浙产金线莲的保肝作用机制。方法用浙产金线莲分别制备水提液和醇提液。选取70只小鼠随机分为正常对照组、模型对照组、水提液和醇提液高、低剂量组和阳性对照组,每组各10只。除正常对照组灌胃生理盐水外,其余组连续灌胃给药1周后,末次给药禁食(不禁水)12 h后,灌胃体积分数为50%乙醇(14 mL·kg^(-1)),建立急性酒精性肝损伤小鼠模型。禁食12 h后摘除小鼠眼球取血,并处死取肝组织。检测血清中ALT、AST活力。制备肝组织的HE病理切片;测定肝组织中丙二醛(MDA)、还原型谷胱甘肽(GSH)含量,酶联免疫法检测肝组织中IL-1β、IL-6的表达。结果小鼠灌胃50%乙醇后造成不同程度的急性酒精性肝损伤。与模型对照组比较,阳性对照组、水提液高剂量组、醇提液高剂量组均能显著降低AST、A LT、MDA(P<0.01),显著升高GSH(P<0.01),能明显下调小鼠肝组织中IL-1β、IL-6表达(P<0.01)。结论水提液和醇提液的保肝作用没有显著差异。浙产金线莲对急性酒精性肝损伤小鼠的保护作用可能与保肝降酶,增强抗氧化能力,降低IL-1β和IL-6表达有关。
OBJECTIVE To compare the effects of water extract and alcohol extract of Anoectochilus roxburghii from Zhejiang on the expression of IL-1β and IL-6 in liver tissue of mice with acute alcoholic liver injury,and to investigate the mechanism of hepatoprotective effect.METHODS Water extract and alcohol extract were prepared from Anoectochilus roxburghii.70 mice were randomly divided into NC group,model group,high and low doses of water extract and alcohol extract groups,and positive control group,with 10 mice in each group.In addition to NC group,the rest groups were given compound continuously for 1 weeks.After 12 hours of fasting,50% ethanol(14 mL·kg^(-1))was given by gavage,and the acute alcoholic liver injury mice model was established.After fasting for 12 hours,the blood was taken by eyeballs removement and liver tissue was taken after death.The activity of ALT and AST in serum was detected.HE pathological sections of liver tissue were prepared.The content of MDA and GSH in liver tissue were measured.The expression of IL-1βand IL-6 in liver tissue were detected by ELISA.RESULTS Acute alcoholic liver injury was induced by intragastric administration of alcohol in mice.Compared with model group,the positive control group,high-dose water extract group and high-dose alcohol extract group could significantly reduce AST,ALT and MDA(P<0.01),significantly increase GSH(P<0.01),and significantly downregulated the expression of IL-1β and IL-6 in mouse liver tissue(P<0.01).CONCLUSION There was no significant difference between water extract and alcohol extract.The protective effect of Anoectochilus roxburghii from Zhejiang on mice with acute alcoholic liver injury may be related to protecting liver,reducing enzyme,enhancing antioxidant capacity and reducing expression of IL-1β and IL-6.
