14种中药材提取物抗聚肌胞苷酸诱导小鼠胸腺萎缩活性的筛选研究 被引量:1

Screening of Anti-Poly(I:C)-induced Mice Thymic Atrophy Activity of 14 Chinese Herbal Medicines Extracts

摘要 目的基于聚肌胞苷酸[Poly(I:C)]诱导的小鼠胸腺萎缩模型对14种中药材的22种提取物进行活性筛选。方法将240只ICR小鼠分为24组,除对照组外的其余各组通过腹腔注射Poly(I:C)生理盐水溶液(隔天注射共3次)造模,各给药组共经口灌胃2周,使末次给药与末次造模在同一天,24 h后处理动物,取胸腺与脾脏称重并计算脏器指数,眼内眦取血进行血细胞分析,胸腺组织进行苏木精-伊红染色以观察病理学变化。结果甘草(内蒙古)组、灵芝组、去壁灵芝孢子粉组以及当归组胸腺指数较模型组显著上升(P<0.05);石斛(普洱)组、去壁灵芝孢子粉组、五倍子组、地黄(山西)组与地黄(禹州)组脾脏指数较模型组有显著升高(P<0.05),当归组、黄芪组有极显著上升(P<0.01);甘草(内蒙古)、去壁灵芝孢子粉、党参组白细胞数较模型组有明显改善(P<0.05);石斛(普洱)组、甘草(内蒙古)组与去壁灵芝孢子粉组较模型组胸腺有显著改善。结论研究发现内蒙古产甘草、灵芝、当归以及去壁灵芝孢子粉对Poly(I:C)诱导小鼠的胸腺萎缩具有积极的改善作用。 OBJECTIVE Activity screening of 22 extracts of 14 Chinese herbal materials based on Poly(I:C)-induced mice thymic atrophy model.METHODS 240 ICR mice were divided into 24 groups.Except the control group,the other groups were injected with Poly(I:C)solution(three times every other day)to establish models.Each group was given intragastric administration of extract for 2 weeks,so that the last administration and the last modeling were on the same day.After 24 hours,the thymus and spleen were weighed and the organ index was calculated.Blood cells were analyzed.The thymus tissue was stained with hematoxylin-eosin to observe the pathological changes.RESULTS The thymus index of Licorice(Inner Mongolia)group,Ganoderma lucidum group,wall removed Ganoderma lucidum spore powder group and Angelica sinensis group was significantly higher than that of model group(P<0.05).The spleen index of Dendrobium(Pu'er)group,wall removed Ganoderma lucidum spore powder group,gallnut group,Rehmannia glutinosa(Shanxi)group and Rehmannia glutinosa(Yuzhou)group was significantly higher than that of model group(P<0.05),and that of Angelica sinensis group and Astragalus membranaceus group was extremely significantly higher than that of model group(P<0.01).The number of white blood cells in group Glycyrrhiza uralensis(Inner Mongolia),Ganoderma lucidum spore powder and Codonopsis pilosula were significantly improved compared with the model group.Compared with the model group,the thymus damage of Dendrobium(Puer)group,Glycyrrhiza uralensis(Inner Mongolia)group and Ganoderma lucidum spore powder group was significantly improved.CONCLUSION The results showed that Licorice(Inner Mongolia),Ganoderma lucidum,Angelica sinensis and wall removed Ganoderma lucidum spore powder have positive effects on Poly(I:C)-induced thymus atrophy in mice.
