摘要
目的检测急性白血病患者血清中可溶性DC-SIGN(the soluble DC-SIGN,sDC-SIGN)的表达水平,分析其在急性白血病诊断、治疗及预后中的临床意义。方法应用酶联免疫吸附试验(ELISA)对67例急性白血病患者和60位健康志愿者血清中sDC-SIGN水平进行检测,比较分析在健康对照组与急性白血病组以及其它分组间的差异,并对20例患者进行了跟踪监测。结果急性白血病患者血清中sDC-SIGN的表达水平明显低于健康对照组,急性淋巴细胞白血病组(ALL)和急性髓细胞白血病组(AML)患者sDC-SIGN表达水平没有显著性差异,而急性髓细胞白血病M2型和M5型患者sDC-SIGN表达水平的差异具有显著性;当sDC-SIGN<2.098μg/mL时,对急性白血病有很高的诊断价值,ROC曲线分析AUC为0.814;患者在诊断或复发状态时sDC-SIGN表达水平低于完全缓解时,对20例患者的连续监测结果显示s DC-SIGN水平持续减低提示病情未得到有效控制,存在复发的危险。结论在急性白血病患者血清中检测到sDC-SIGN的表达并随病情发展而变化,可以作为急性白血病的诊断、病情判断和预后评估的新的分子标志物。
Objective To detect the levels of sDC-SIGN(the suloble DC-SIGN)expression in serum of acute leukemia patients,analyze its clinical significance in the diagnosis,treatment and prognosis of acute leukemia.Methods The expression of sDC-SING in serum from 67 acute leukemia patients and 60 healthy controls was detected by enzyme-linked immunosorbent assay(ELISA),compare to the levels of sDC-SIGN in the different groups and 20 patients were monitored.Results The level of sDC-SIGN was lower in acute leukemia patients than in the healthy controls.There was no difference in the expression of sDC-SIGN between ALL and AML patients,and significant differences between M2 and M5 patients.As the sDC-SIGN level of<2.098μg/mL,the value in the diagnosis of acute leukemia was high,AUC of ROC curve analysis is 0.814.The level of sDC-SIGN in serum in first-diagnosis/relapse patients was lower than in complete remission patients.20 cases of acute leukemia patients were monitored during the test,the continuous decrease of the sDC-SIGN level indicated the risk of recurrence.Conclusion The sDC-SIGN are first detected in the serum of patients with acute leukemia can be used as a new molecular marker for diagnosis,observation variety state and prognosis evaluation of acute leukemia.
