摘要
目的:对生姜不同极性部位的止呕作用进行比较研究,找到最佳药效部位和给药剂量,并分析最佳药效部位的化学成分。方法:采用经典大鼠异嗜高岭土模型,不同极性部位分为高、中、低3个剂量给药,运用UPLC-QTrap-MS/MS联合衍生化法测定回肠和延髓中的五羟色胺及延髓中的多巴胺含量,酶联免疫吸附法检测血浆中胃动素、胆囊收缩素、降钙基因相关肽的含量,并利用UPLC-ESI-Q-TOF-MS/MS对筛选出的最佳药效部位进行化学成分鉴定。结果:B段(经MCI柱色谱后,10%~30%甲醇水的洗脱成分)中剂量组大鼠的异嗜行为得到极显著改善(P<0.01),各项指标与模型组大鼠相比差异极显著(P<0.01)。在B段共鉴定出44个化合物,其中有25个糖苷类化合物。结论:B段给药剂量为7 mg·kg^(-1)时止呕效果最好,即生姜中极性较大的成分发挥着更好的药效作用,可能对外周系统与中枢系统均有作用,其中糖苷类化合物很可能是其主要药效成分。
Objective: The antiemetic effects of different polar parts of ginger were compared, the most effective parts and dosage were identified, and the chemical components of the most effective parts were analyzed.Methods: The classical rat model of heterophilia of kaolin was applied, given different polar parts and divided into three dose groups. The contents of serotonin in ileum and medulla oblongata, as well as dopamine levels in medulla oblongata were determined by UPLC-QTrap-MS/MS combined derivatization method. The expression levels of motilin,cholecystokinin and calcium gene in plasma were detected by enzyme-linked immunosorbent assay. UPLC-ESI-QTOF-MS/MS was used to analyze the chemical constituents of the selected best effective parts.Results:The xenophilic behavior of rats in fraction B medium-dose group was significantly ameliorated(P< 0. 01), and the differences in various indicators were the most significant as compared with those in the model group(P< 0. 01). A total of 44compounds were identified in fraction B, including 25 glycosides.Conclusion: The emesis-stopping effect was the best when the dose of fraction B was 7 mg·kg^(-1), that is, the high-polar components in fresh ginger played the most significant pharmacodynamic role, which might affect both peripheral system and central system. The glycosides are likely to be one of the main pharmacodynamic components.
