miR-155在结肠癌组织中的表达及其对HCT116、SW480细胞增殖与凋亡的影响

摘要 目的 分析结肠癌组织中微小RNA-155(miR-155)表达,并明确其对HCT116、SW480细胞增殖与凋亡的影响。方法 收集2020年6月—2021年6月盐城市第三人民医院内镜中心结肠镜检查病理标本提示为结肠癌,且符合入组条件的78例结肠癌活检组织使用qRT-PCR检测miR-155的表达情况。体外培养人结肠癌HCT116、SW480细胞,予以miR-155抑制剂处理后,观察细胞增殖率和凋亡率的变化。结果 结肠癌组织miR-155水平(2.41±0.12)、癌旁组织(1.01±0.09),结肠癌组织miR-155水平较高,差异有统计学意义(t=82.430,P<0.05);miR-155抑制剂能有效降低HCT116、SW480细胞增殖速度,差异有统计学意义(P<0.05);miR-155抑制剂作用组HCT116、SW480细胞凋亡率明显增加,差异有统计学意义(t=47.983、42.506,P<0.05)。结论 miR-155在结肠癌组织中呈现高表达状态,对其进行抑制能够降低HCT116、SW480细胞增殖率,促进HCT116、SW480细胞凋亡,其有望能成为结肠癌患者治疗靶点,以期能够改善患者预后。 Objective To analyze the expression of microRNA-155(miR-155) in colon cancer tissue,and to clarify its effect on the proliferation and apoptosis of HCT116 and SW480 cells.Methods From June 2020 to June 2021,colonoscopy pathological specimens from the Endoscopy Center of the Third People’s Hospital of Yancheng City were collected to indicate colon cancer.And 78 cases of colon cancer biopsy tissue that met the inclusion conditions were detected by qRT-PCR for the expression of miR-155.Human colon cancer HCT116 and SW480 cells were cultured in vitro and treated with miR-155 inhibitor to observe the changes of cell proliferation rate and apoptosis rate.Results The levels of miR-155 in colon cancer tissues(2.41±0.12) and in adjacent tissues(1.01±0.09) were higher in colon cancer tissues,and the difference was statistically significant(t=82.430,P<0.05);miR-155 inhibitor could effectively reduce the proliferation rate of HCT116 and SW480 cells,the difference was statistically significant(P<0.05);The apoptosis rate of HCT116 and SW480 cells in miR-155 inhibitor group increased significantly,the difference was statistically significant(t=47.983,42.506,P<0.05).Conclusion miR-155 is highly expressed in colon cancer tissues,and its inhibition can reduce the proliferation rate of HCT116 and SW480 cells and promote the apoptosis of HCT116 and SW480 cells.It is expected to become a therapeutic target for patients with colon cancer,in order to improve the prognosis of patients.
