离子色谱法同时测定乌梅中4种有机酸的含量 被引量:9

摘要 目的:建立离子色谱法同时测定乌梅药材及中药配方颗粒中4种有机酸成分(L-苹果酸、草酸、延胡索酸和柠檬酸)的含量。方法:样品经处理后用分离柱分离,以KOH为淋洗液在4~54 mmol/L的浓度下梯度洗脱,抑制电流为75 mA。结果:在优化的梯度条件下,4种有机酸均能基线分离,且待测物浓度与其峰面积在一定范围内呈良好线性,重复性、稳定性以及精密度的相对标准偏差RSD均小于3.0%,加标回收率为98%~100%。结论:该方法同时测定乌梅药材及中药配方颗粒多种有机酸含量,操作简便、准确、快速,结合药理分离提取有效的有机酸成分,具有提高乌梅的使用效率以及研究阐明乌梅有机酸成分药理作用的机制等实际意义;同时该方法中L-苹果酸、草酸、延胡索酸和柠檬酸四种有机酸分离度良好,适合于快速测定乌梅、青梅等富含有机酸的药材及食材,能够为进一步开发乌梅等富含有机酸的药材及食材提供可靠的科学依据。 Objective:To establish an ion chromatography method for the simultaneous determination of four organic acids in fructus mume herbs and Chinese herbal formula granules:malic acid,oxalic acid,fumaric acid and citric acid.Methods:After treatment,the samples were separated by a separation column,and then eluted with KOH at a concentration of 4-54 mmol/L with a gradient elution current of 75 mA.Results:Under the optimized gradient conditions,the four organic acids could be separated from the baseline,and the concentration of the measured compounds was linear with its peak area in a certain range.The RSDS of repeatability,stability and precision were all less than3.0%,and the recoveries were 98%-100%.Conclusions:The method is simple,accurate and rapid for simultaneous determination of various organic acids in fructus mume medicinal materials and Chinese medicine formula granules.Combined with pharmacological separation and extraction of effective organic acids,it has practical significance to improve the use efficiency of fructus mume and to study and clarify the pharmacological mechanism of organic acids in fructus mume.At the same time,the separation of L-malic acid,oxalic acid,fumaric acid and citric acid in this method is good,which is suitable for the rapid determination of organic acid-rich medicinal materials and food materials such as fructus mume and green plum,and can provide a reliable scientific basis for the further development of organic acid-rich medicinal materials and food materials such as fructus mume.
