摘要
目的观察调胃承气汤对脑出血急性期大鼠胃肠组织SCF/c-kit信号通路的影响,探讨其调节胃肠动力障碍的作用机制。方法选取40只健康Wistar大鼠,按照随机数字法将其分为假手术组、模型组、西药组和中药组,每组10只。除假手术组只进针不注射外,其他组均采用自体血注入法建立脑出血大鼠模型。成功造模后,中药组和西药组分别给予113.4 mg/mL的调胃承气汤悬浊液和0.135 mg/mL的莫沙必利悬浊液10 mL/(kg·d)灌胃,假手术组和模型组予以等容量生理盐水灌胃,均连续灌胃7 d。分别采用Western blot法和RT-PCR法检测大鼠胃、小肠、大肠、盲肠组织中c-kit、SCF蛋白及mRNA表达情况。结果模型组大鼠胃肠各组织中的c-kit、SCF蛋白及mRNA相对表达量均明显低于假手术组(P均<0.05);中药组和西药组大鼠胃肠各组织中的c-kit、SCF蛋白及mRNA相对表达量均明显高于模型组(P均<0.05),中药组和西药组各指标比较差异均无统计学意义(P均>0.05),但中药组各指标有高于西药组的趋势。结论调胃承气汤改善脑出血急性期胃肠动力障碍的作用机制与提高SCF/c-kit信号通路相关蛋白及mRNA的表达有关。
Objective It is to observe the effect of Tiaowei Chengqi Decoction on SCF/c-kit signaling pathway in gastrointestinal tissue of rats with intracerebral hemorrhage at acute stage,and to explore the mechanism of regulating gastrointestinal motility disorder.Methods Forty healthy Wistar rats were selected and divided into sham operation group,model group,western medicine group and Chinese medicine group according to random number method,with 10 rats in each group.Except for the sham-operated group,which was only injected with needles but not injected,the other groups were all used autologous blood injection to establish rat models of cerebral hemorrhage.After successful modeling,the traditional Chinese medicine group and the western medicine group were respectively given 113.4 mg/mL Tiaowei Chengqi Decoction suspension and 0.135mg/mL mosapide suspension 10 mL/(kg·d)by gavage,while the sham operation group and model group were given equal volume normal saline by gavage.After feeding for 7 days,the expression levels of SCF,c-kit protein and mRNA in the stomach,small intestine,large intestine and cecum were detected by Western Blot and RT-PCR respectively.Results Compared with sham operation group,the expression levels of SCF,c-kit protein and mRNA in gastrointestinal tissues of model group were significantly decreased(all P<0.05).Compared with the model group,the above indexes in the traditional Chinese medicine group and the western medicine group were significantly increased(all P<0.05).There was no significant difference in the indexes between the traditional Chinese medicine group and the western medicine group(all P>0.05),but the indexes in the traditional Chinese medicine group were higher than those in the western medicine group.Conclusion The mechanism of Tiaowei Chengqi Decoction in improving gastrointestinal motility disorder in cerebral hemorrhage at acute stage is related to increase the related proteins and mRNA of SCF/c-kit signaling pathway.
