精原干细胞诱导方法与机理研究进展

摘要 雄性生殖干细胞(spermatogonial stem cells,SSCs)是一类具备自我更新和分化潜能的干细胞,也是雄性个体一生中不断产生精子细胞的基础。雄性生殖干细胞的体外诱导分化能为精子发生机理的研究提供重要理论依据,对恢复不育个体的精子发生,医治雄性动物的生殖障碍具有重要意义。该文主要对精原干细胞诱导分化的试剂诱导法和细胞移植法两种方法,以及细胞因子诱导、转录因子诱导、miRNA诱导等三种诱导机理进行综述,旨在对雄性动物辅助生殖的研究提供借鉴。 The male reproductive stem cells(spermatogonial stem cells,SSCs)are a kind of stem cells with the potential of self-renew and differentiation,and are also the basis of the continuous production of sperm cells in male individuals throughout their lives.In vitro induced differentiation of reproductive stem cells provides an important theoretical basis for the research of spermatogenesis mechanism,which is of great significance for the reconstruction of spermatogenesis in sterile individuals and the treatment of male reproductive barriers.In this paper,two methods of spermatogonial stem cell differentiation induction,reagents induction and cell transplantation,as well as three induction mechanisms,namely,cytokine induction,transcription factor induction and miRNA induction,are reviewed in order to provide reference for the study of assisted reproduction in male animals.
