复方苦参注射液通过保护线粒体防治大鼠急性放射性皮炎的研究 被引量:7

摘要 目的:探讨复方苦参注射液治疗大鼠放射性皮炎的作用以及可能机制。方法:将30只SD大鼠随机分为空白对照组、放疗模型组、皮肤给药组、腹腔给药组和联合治疗组。除空白对照组外,其余大鼠右侧大腿皮肤处给予4 mV电子线单次30 Gy照射,建立急性放射性皮炎模型。自照射后第2天起开始治疗,皮肤给药组予以复方苦参注射液湿敷照射区域皮肤,每日2次,每次10 min;腹腔给药组予以0.3 mL/100 g剂量复方苦参注射液进行腹腔注射,每日1次;联合治疗组同时予以复方苦参注射液皮肤湿敷和腹腔给药;空白对照组、放疗模型组予以等量生理盐水腹腔注射和患处湿敷。连续处理14 d,观察各组大鼠皮肤改变情况。照射14 d后处死大鼠,光镜和电镜观察患处皮肤组织改变,ELISA法检测皮肤组织IL-6、TNF-α及线粒体延胡索酸酶、顺乌头酸酶、8-羟基脱氧鸟苷(8-OHDG)的表达量。结果:相对于放疗模型组,应用复方苦参注射液治疗能明显改善大鼠皮肤组织及线粒体结构损伤,减少皮肤组织IL-6和TNF-α含量,提高线粒体顺乌头酸酶和延胡索酸酶活性,降低组织8-羟基脱氧鸟苷含量;效果以腹腔给药组最好,与放疗模型组相比差异具有统计学意义(P <0.05)。结论:复方苦参注射液能防治大鼠急性放射性皮炎,其机制可能与保护线粒体的结构和酶的功能相关。 Objective:To explore the effect of compound Kushen Injection on radiation dermatitis in rats and its possible mechanism.Methods:30 SD rats were randomly divided into the blank control group,the radiotherapy model group,the skin administration group,the intraperitoneal administration group and the combination treatment group.Acute radiation dermatitis model was established by irradiation of a single 30 Gy of 4 mV electron beam to the skin of the right thigh in the rats except the blank control ones.The second day after the radiation,the blank control group and the radiotherapy model group were given intraperitoneal injection of normal saline,the skin administration group was treated with local skin hydropathic compress of compound Kushen Injection(twice a day,10 minutes each time),the intraperitoneal administration group treated with intraperitoneal injection of compound Kushen Injection(0.3 mL/100 g,once a day),and the combination treatment group was treated with both local skin hydropathic compress and intraperitoneal injection.The treatment was continued for 14 days,and the skin changes of the rats were observed in each group.Skin tissues changes were observed under light microscope and electron microscope.The expressions of IL-6 and TNF-α in skin tissues were detected by ELISA,and the activities and the contents of aconitase,fumarase and 8-hydroxydeoxybird(8-OHDG)in mitochondria were detected by the corresponding kit.Results:Compared with those in the radiotherapy model group,the damage of the skin tissues and the mitochondrial structure were improved,the contents of IL-6 and TNF-α were reduced,the activities of aconitase and fumarase in the mitochondrial were enhanced,and the content of 8-OHDG was reduced by the intervention of compound Kushen Injection;the effect was best in the intraperitoneal administration group,which was significantly different from the radiotherapy model group(P < 0.05).Conclusion:The mechanism of compound Kushen Injection in the treatment of acute radiation dermatitis in rats may be related to the protection of the structure of mitochondria and the function of enzymes.
