花生组织抗氧化生理生化指标快速测定方法

摘要 为了提高基础生理生化试验过程中的效率,本文基于多次试验和分析,优化了传统抗氧化生理生化指标的测定方法。利用干旱胁迫处理的花生根系和叶片为试验材料,通过测定超氧化物歧化酶(SOD)、过氧化物酶(POD)、丙二醛(MDA)、可溶性蛋白(SP)等指标,验证优化后的快速测定方法与传统测定方法测定结果的吻合度。结果表明,快速测定方法仅用微量样本和传统方法测定所需的一半时间;所获数据与传统测定方法无显著差异。大批量样本生理生化指标测定时,可利用此快速测定方法代替传统方法,有效解决了目前存在的样本用量大、研磨不均一、耗时长、误差偏大等问题。 In order to improve efficiency of fundamental test determinations, traditional antioxidant physiological and biochemical index determination method was optimized. In order to verify results derived from optimized rapid determination method and traditional determination method, superoxide dismutase(SOD), peroxidase dismutase(POD), malondialdehyde(MDA), soluble protein(SP) and other indicators were processed by using peanut roots and leaves treated with drought stress. The results showed that the rapid determination method only had employed a trace of samples and consumed half the time compared to traditional method. Therefore, there was no significant difference from the data obtained by the traditional determination method. When measuring the physiological and biochemical indicators of a large scale of samples under adversity stress, rapid measurement method can be served as alternative choice of traditional measurement method, which has effectively solved the current problems of large sample volume, uneven grinding, time-consuming, and big errors, thereby improving experiment efficiency.
