变异系数法-AHP综合加权结合响应面法优选板蓝根配方颗粒提取工艺 被引量:6

Extraction Process Optimization for Isatidis Radix Formula Granules by Variability Coefficient Method-AHP Comprehensive Weighting Combined with Response Surface Methodology

摘要 目的:优选板蓝根配方颗粒的最佳提取工艺。方法:以煎煮时间、加水量、煎煮次数为考察因素,利用Box⁃Behnken响应面模型,设计三因素三水平的响应面试验,通过变异系数法结合层次分析法(AHP)对板蓝根出膏率、尿苷、腺苷、鸟苷和(R,S)⁃告依春的含量分别赋予权重,并以综合加权评分值为评价指标,考察3个因素对板蓝根提取效果的影响,优选最佳提取工艺,并进行工艺验证。结果:板蓝根配方颗粒的最佳提取工艺为加12倍量水,煎煮3次,每次40 min,验证试验的综合评分分别为0.984、0.971、0.963,各质量指标的RSD均小于5.0%。结论:优选的提取工艺稳定、简单、可行,可为板蓝根配方颗粒的制剂工艺研究提供参考。 Objective:To optimize the extraction process of Isatidis Radix formula granules.Methods:Taking the time of decoction,amount of water added and decoction times as the influencing factors,Box-Behnken response surface model was used to design a response surface test with the three factors at three levels.Coefficient of variation method combined with analytic hierarchy process(AHP)was used to weight the extract rate of Isatidis Radix and the contents of uridine,adenosine,guanosine and(R,S)-Gaoyichun,and the comprehensive weighted score was used as the evaluation index.The influences of the three factors on the extraction effect of Isatidis Radix were investigated,and the extraction process was optimized and verified.Results:The best extraction process was as follows:adding 12-fold amount of water and decocting 3 times with 40 min for each time.The average score of the three groups of verification tests was 0.973,and the RSD value was less than 5.0%.Conclusion:The preferred extraction process is stable,simple and feasible,and can provide reference for the research on the preparation process of Isatidis Radix formula granules.
