摘要
目的 研究单核细胞P糖蛋白(Pgp)、多药耐药相关蛋白(MRP)在肺结核病患者外周血中的表达情况,探讨其与多药耐药的相关性。方法 回顾性分析144例肺结核病患者临床资料及同期50例健康体检者资料,采用Trizol法检测Pgp、MRP基因在血清样本中表达情况,根据药敏试验将144例肺结核病患者分为耐药组和初治肺结核病组,分析Pgp、MRP基因在肺结核患者血清中的表达情况,探讨肺结核病患者耐药的影响因素。结果 Pgp、MRP在肺结核病组表达量及阳性率均高于对照组,耐药组Pgp、MRP表达量及阳性率均高于初治结核病组,差异均有统计学意义(P<0.05)。年龄、居住地、Pgp、MRP表达量是肺结核病患者耐药的主要影响因素。结论 Pgp、MRP在肺结核病患者呈高表达,是肺结核患者耐药的独立影响因素。
Objective To study the expression of P-glycoprotein(Pgp)and multidrug resistance associated protein(MRP)in peripheral blood of patients with pulmonary tuberculosis,and to explore the correlation between Pgp and multidrug resistance.Methods The clinical data of 144patients with pulmonary tuberculosis were retrospectively analyzed.The expression of Pgp and MRP genes in serum samples was detected by Trizol method.According to drug sensitivity test,144patients with pulmonary tuberculosis were divided into drug resistance group and newly treated group.The expression of Pgp and MRP genes in serum of patients with pulmonary tuberculosis was analyzed,and the influencing factors of drug resistance of patients with pulmonary tuberculosis were discussed.Results The expression and positive rate of PGP and MRP in the tuberculosis group were higher than those in the control group,and the expression and positive rate of PGP and MRP in the drug-resistant group were higher than those in the newly treated tuberculosis group(P<0.05).Age,rural area,expression of PGP and MRP were the main influencing factors of drug resistance.Conclusion The expression of PGP and MRP is high in patients with pulmonary tuberculosis,which is an independent factor of drug resistance in patients with pulmonary tuberculosis,and it may be the target gene of drug resistance prediction in patients with pulmonary tuberculosis.
