姜黄素通过NLRP3炎症小体通路抑制AngⅡ诱导的子宫内膜间质细胞的分泌活性 被引量:2

Inhibition of curcumin on AngⅡinduced secretory activity of endometrial stromal cells through NLRP3 inflammasome pathway

摘要 目的研究姜黄素对血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)诱导的子宫内膜间质细胞(endometrial stromal cells,ESCs)增殖、分泌活性以及细胞内NLRP3炎症小体激活的影响。方法获取正常生育期女性的子宫内膜,消化、分离及鉴定内膜里的ESCs。检测一定浓度和时间范围内的AngⅡ对ESCs的增殖情况,选择最佳的AngⅡ作用浓度和时间做后续实验。细胞设为正常组、姜黄素组、AngⅡ组以及AngⅡ+姜黄素组4组。ELISA法检测各组细胞培养悬液中Ⅰ型胶原(typeⅠcollagen,ColⅠ)和纤黏连蛋白(fibronectin,FN)的含量;蛋白免疫印迹技术评估ESCs中NLRP3、caspase-1和IL-1β的含量。结果与正常组相比,一定浓度和时间范围内AngⅡ均能促进ESCs的增殖(均P<0.05),并且浓度为10^(-6)mol/L AngⅡ作用48 h时ESCs的增殖(0.1228±0.0013)效果最明显(P<0.01);姜黄素组与正常组相比,ColⅠ和FN的含量、炎性小体(NLRP3、caspase-1和IL-1β)的含量差异均无统计学意义(均P>0.05)。与正常组相比,AngⅡ组的ColⅠ(0.7966±0.0839)和FN(0.6461±0.0603)的含量、NLRP3(0.573±0.039)、caspase-1(1.135±0.050)和IL-1β(1.291±0.046)的含量均明显增加(均P<0.05);与AngⅡ组相比,AngⅡ+姜黄素组的ColⅠ(0.5706±0.0631)和FN(0.5027±0.0428)的含量、NLRP3(0.344±0.036)、caspase-1(0.685±0.035)和IL-1β(0.714±0.039)的含量均明显降低(均P<0.05)。结论姜黄素能够明显拮抗AngⅡ诱导的ESCs的增殖能力和分泌活性,这与降低细胞内NLRP3炎症小体及IL-1β的表达密切相关。 Objective To study the effects of curcumin on the proliferation and secretory activity of endometrial stromal cells(ESCs)induced by angiotensinⅡ(AngⅡ)and the activation of intracellular NLRP3 inflammatory bodies.Methods Endometrium of women in normal reproductive period was obtained,and ESCs in endometrium were digested,isolated and identified.The proliferation of ESCs by induced AngⅡin a certain concentration and time range was detected,and the best concentration and time of AngⅡwere selected for follow-up experiments.The cells were designed into four groups:normal group,curcumin group,AngⅡgroup and AngⅡ+curcumin group.The contents of typeⅠcollagen(ColⅠ)and fibronectin(FN)in cell culture suspension were detected by ELISA.The content of of NLRP3,caspase-1 and IL-1βin ESCs were evaluated by Western blot.Results Compared with the normal group,AngⅡcould promote the proliferation of ESCs in a certain concentration and time range(all P<0.05),and the proliferation effect of ESCs was the most obvious(0.1228±0.0013)at the concentration of 10^(-6)mol/L for 48 hours(P<0.01).There were no statistically significant difference in the contents of ColⅠFN,NLRP3,caspase-1 and IL-1βbetween the normal group and the curcumin group(all P>0.05).Compared with the normal group,the contents of ColⅠ(0.7966±0.0839)and FN(0.6461±0.0603),NLRP3(0.573±0.039),caspase-1(1.135±0.050)and IL-1β(1.291±0.046)in AngⅡgroup increased significantly(all P<0.05).Compared with AngⅡgroup,the contents of ColⅠ(0.5706±0.0631),FN(0.5027±0.0428),NLRP3(0.344±0.036),caspase-1(0.685±0.035)and IL-1β(0.714±0.039)in AngⅡ+curcumin group decreased significantly(all P<0.05).Conclusion Curcumin can significantly antagonize the proliferation and secretory activity of ESCs induced by AngⅡ,which is closely related to the reduction of NLRP3 inflammatory body and IL-1βin cells.
