当归多糖对复发性流产大鼠的干预效果和作用机制研究 被引量:8

Intervention effect of Angelica sinensis polysaccharide on rats with recurrent spontaneous abortion and study on the mechanism

摘要 目的观察当归多糖对复发性流产(RSA)大鼠的干预效果,分析作用机制。方法选取34只健康的雌性CBA/J大鼠、13只健康的雄性DBA/2大鼠及4只健康的雄性BALB/c大鼠。合笼后,8只正常妊娠大鼠为正常组,剩余26只雌性大鼠建立RSA大鼠模型,将成功妊娠的24只大鼠随机分为模型组、低剂量当归多糖组及高剂量当归多糖组,每组8只。计算大鼠流产率。应用放射免疫法检测血清卵泡刺激素(FSH)、黄体生成素(LH)、催乳素(PRL)及雌二醇(E_(2))水平。应用酶联免疫法检测血浆凝血酶原时间(PT)和纤维蛋白原(Fbg)水平。采用免疫印迹法检测磷脂酰肌醇3激酶(PI3K)、蛋白激酶B(AKT)蛋白水平。结果正常组、模型组、低当归多糖组及高当归多糖组大鼠流产率分别为(3.69±0.26)%、(68.98±16.29)%、(42.59±15.59)%及(25.13±13.29)%,4组比较差异有统计学意义(F=17.265,P=0.001)。模型组、低当归多糖组及高当归多糖组大鼠流产率均显著高于正常组(t=11.330,P=0.001;t=7.056,P=0.001;t=4.562,P=0.001);低当归多糖组、高当归多糖组大鼠流产率均显著低于模型组(t=3.331,P=0.005;t=5.899,P=0.001);高当归多糖组大鼠流产率显著低于低当归多糖组(t=2.411,P=0.030)。各组大鼠血清FSH、LH、PRL及E_(2)水平,PT,Fbg水平,P13K和AKT蛋白水平比较差异均有统计学意义(均P<0.05)。结论当归多糖可以改善RSA大鼠激素水平和凝血指标,降低流产率,作用机制可能与PI3K/AKT信号通路蛋白有关。 Objective To observe the intervention effect of Angelica sinensis polysaccharide on rats with recurrent spontaneous abortion(RSA),analyze the mechanism.Methods Thirty-four healthy female CBA/J rats,thirteen healthy male DBA/2 rats,and four healthy male BALB/c rats were selected,after caging,eight normal pregnant rats were designed as normal group,the other 26 female rats were collected to establish RSA rat model,then 24 pregnant rats were randomly divided into model group,low-dose Angelica sinensis polysaccharide group,and high-dose Angelica sinensis polysaccharide group,8 rats in each group.The abortion rats were calculated.Radioimmunoassay was used to detect the levels of serum follicle-stimulating hormone(FSH),luteinizing hormone(LH),prolactin(PRL),and estradiol(E_(2)).Enzyme-linked immunosorbent assay was used to detect plasma prothrombin time(PT)and fibrinogen(Fbg).Western blotting was used to detect the levels of phosphatidylinositol 3 kinase(PI3 K)and protein kinase(AKT)proteins.Results The abortion rates in normal group,model group,low-dose Angelica sinensis polysaccharide group,and high-dose Angelica sinensis polysaccharide group were(3.69±0.26)%,(68.98±16.29)%,(42.59±15.59)%,and(25.13±13.29)%,respectively,there was statistically significant difference(F=17.265,P=0.001).The abortion rates in model group,low-dose Angelica sinensis polysaccharide group,and high-dose Angelica sinensis polysaccharide group were statistically significantly higher than that in normal group(t=11.330,P=0.001;t=7.056,P=0.001;t=4.562,P=0.001);the abortion rates in low-dose Angelica sinensis polysaccharide group and high-dose Angelica sinensis polysaccharide group were statistically significantly lower than that in model group(t=3.331,P=0.005;t=5.899,P=0.001);the abortion rate in high-dose Angelica sinensis polysaccharide group was statistically significantly lower than that in low-dose Angelica sinensis polysaccharide group(t=2.411,P=0.030).There were statistically significant differences in serum levels of FSH,LH,PRL,and E_(2),PT,Fbg level,levels of P13 K and AKT proteins among different groups(P<0.05).Conclusion Angelica sinensis polysaccharide can improve hormones levels and coagulation indicators of RSA rats,reduce abortion rate,the mechanism may be related to proteins of P13 K/AKT signaling pathway.
