霍山石斛通过激活自噬和抑制凋亡促进脱发模型小鼠生发作用 被引量:4

Dendrobium huoshanense promotes hair growth in mouse model of alopecia by activating autophagy and inhibiting apoptosis

摘要 文章以C57BL/6J小鼠为研究对象,构建脱发小鼠模型来研究霍山石斛对脱发的治疗作用及机制。将C57BL/6J小鼠随机分为对照组、霍山石斛组(涂抹石斛汁),实验结束后对比小鼠毛发生长情况,取背部皮肤进行H&E染色、免疫组化染色,通过Western Blot和实时荧光定量聚合酶链式反应(polymerase chain reaction,PCR)检测皮肤组织中B淋巴细胞瘤-2(Bcl-2)、p62、微管相关蛋白1轻链3-β(LC3Ⅱ)和转换生长因子-β1(TGF-β1)的蛋白和mRNA的表达情况。结果表明,霍山石斛能够促进毛发生长,改善皮肤毛囊形态,上调自噬关键因子LC3Ⅱ和抑制凋亡分子Bcl-2的表达,下调转化生长因子TGF-β1的表达,表明霍山石斛通过激活自噬和抑制凋亡缓解脱发。 In this study,C57BL/6J mice(6-8 weeks old)were used to establish the mouse model of alopecia to explore the therapeutic effect and mechanism of Dendrobium huoshanense on alopecia.C57BL/6J mice were randomly divided into a control group and a Dendrobium huoshanense group.After the experiment,the hair growth of mice was compared,the H&E staining and immunohistochemical staining of back skin sections were conducted,the protein and mRNA expression of B-cell lymphoma-2(Bcl-2),p62,microtubule-associated protein 1 light chain 3-β(LC3Ⅱ)and transforming growth factor-β1(TGF-β1)in skin tissue were further determined by Western Blot and real-time quantitative polymerase chain reaction(PCR).The results indicate that Dendrobium huoshanense can promote the hair growth,improve the hair follicle morphology,up-regulate the expression of autophagosome marker LC3Ⅱand anti-apoptotic molecule Bcl-2 and inhibit the expression of TGF-β1,showing that Dendrobium huoshanense relieves the hair loss by activating autophagy pathway and inhibiting apoptosis.
