摘要
目的:建立超高效液相色谱-串联质谱法(Ultra Performance Liquid Chromatography-tandem Mass Spectrometry,UPLC-MS/MS)同时测定淡水养殖用水中25种抗生素的检测方法。方法:500 mL淡水养殖用水用盐酸溶液调节pH至2.5,试样经HLB萃取柱富集,甲醇洗脱,经过40 ℃空气吹干后定容,采用超高效液相色谱-串联质谱法检测,内标法定量。结果:25种抗生素在0~50.0 ng/L线性关系良好,相关系数为0.990 61~0.999 83,检出限为0.1~1.0 ng/L,磺胺类药物平均回收率为100.7%~115.4%,RSD为1.05%~4.69%,喹诺酮类药物平均回收率为90.1%~111.0%,RSD为2.20%~9.55%。结论:本方法快速、稳定、灵敏度和准确度高,适用于淡水养殖用水中25种磺胺类和喹诺酮类抗生素残留检测。
Objective:To establish a method for the simultaneous determination of 25 kinds of antibiotics in freshwater aquaculture water by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS).Method:The 500 mL of freshwater aquaculture water was adjusted to pH 2.5 with hydrochloric acid solution.The sample was enriched by HLB extraction column,eluted with methanol,dried in air at 40 ℃,then volume.detected by ultra performance liquid chromatography-tandem mass spectrometry,and the contents were quantified by internal Standard Method.Result:the linear relationship of 25 antibiotics was good in the range of 0~50.0 ng/L,the correlation coefficient was 0.990 61~0.999 83,the detection limit was 0.1~1.0 ng/L,the average recovery of sulfonamides was 100.7%~115.4%,RSD was 1.05%~4.69%,and the average recovery of quinolones was 90.1%~111.0%,RSD was 2.20%~9.55%.Conclusion:the method is rapid,stable,sensitive and accurate.It is suitable for the determination of 25 sulfonamides and quinolones residues in freshwater aquaculture water.
