摘要
目的探讨菊苣酸对口腔癌KB细胞增殖、侵袭与黏附的影响及机制。方法将KB细胞分为对照组和低、中、高剂量实验组,分别用0,5,10和20μmol·L^(-1)菊苣酸干预24 h。比较4组细胞的穿膜细胞数、细胞黏附率,以及磷脂酰肌醇3激酶p85亚单位(PI3Kp85)和磷酸化蛋白激酶B(p-Akt)的表达情况。结果低、高剂量实验组和对照组的穿膜细胞数百分比分别为(79.37±8.04)%,(64.66±7.61)%和(100.00±0)%,细胞黏附率分别为(84.06±3.27)%,(64.64±7.22)%和(100.00±0)%,PI3Kp85相对表达量分别为0.70±0.10,0.08±0.02和0.78±0.05,p-Akt相对表达量分别为0.50±0.06,0.27±0.05和0.81±0.07,低、高剂量实验组的上述指标与对照组比较,差异均有统计学意义(P<0.05或P<0.01)。结论菊苣酸能够抑制口腔癌KB细胞的增殖、侵袭与黏附,该作用与抑制PI3K/Akt信号通路的活化有关。
Objective To investigate the effect and mechanism of chicory acid on the proliferation,invasion and adhesion of oral carcinoma KB cells.Methods The KB cells were divided into control group and experimental-L,experimental-M and experimental-H groups,and treated with 0,5,10 and 20μmol·L^(-1) chicoric acid for 24 h,respectively.The number of transmembrane cells,cell adhesion rate,phosphatidylinositol 3 kinase p85 subunit(PI3 Kp85)and phosphorylated protein kinase B(p-Akt)expression in 4 groups was compared.Results The percentages of transmembrane cells in the experimental-L,experimental-H and control groups were(79.37±8.04)%,(64.66±7.61)%and(100.00±0)%,the cell adhesion rates were(84.06±3.27)%,(64.64±7.22)%and(100.00±0)%,the relative expression levels of PI3 Kp85 were 0.70±0.10,0.08±0.02 and 0.78±0.05,the relative expression levels of p-Akt were 0.50±0.06,0.27±0.05 and 0.81±0.07,respectively.Compared with the control group,the above indicators of experimental-L and experimental-H groups showed statistically significant differences(P<0.05 or P<0.01).Conclusion Chicory acid can inhibit the proliferation,invasion and adhesion of oral carcinoma KB cells,which is related to the blocking of the activation of PI3K/Akt signaling pathway.
