非洲猪瘟病毒感染致母猪卵巢组织损伤的病理学观察 被引量:2

摘要 为明确非洲猪瘟病毒(ASFV)感染对母猪卵巢组织的损伤作用,本研究分离了健康母猪卵巢中原代颗粒细胞(GCs),采集了ASFV感染的母猪卵巢组织。采用荧光定量PCR方法检测ASFV的复制情况以及感染ASFV的卵巢组织中炎症相关因子的转录情况;利用苏木精-伊红(HE)染色和免疫组织化学(IHC)技术分析感染后的卵巢组织病理变化和促卵泡刺激素受体(FSHR)及促黄体激素受体(LHR)的免疫组化情况。结果显示,ASFV能感染GCs且大量复制,与对照组相比,ASFV感染卵巢组织中炎症相关因子转录水平显著升高,卵巢黄体和卵巢组织间质大面积淤血,淋巴细胞浸润,黄体细胞空泡化变性;卵泡内坏死细胞周围有少量巨噬细胞浸润。ASFV感染导致FSHR和LHR转录水平显著降低,IHC结果显示,ASFV感染引起卵巢中FSHR及LHR阳性细胞密度显著降低。本试验结果为研究ASFV感染引起母猪生殖系统病变及其繁殖障碍提供了参考,积累了数据。 In order to clarify the damage effect on sows ovarian tissue after ASFV infection,primary granulosa cells(GCs)were isolated from healthy sow ovaries,and ovaries samples were collected ASFV infection.Replication of ASFV and the transcription of inflammatory related factors in ovarian tissues infected with ASFV were detected by fluorescence quantitative PCR.Hematoxylin eosin staining(HE)and immunohistochemistry(IHC)were used to analyze the pathological changes of ovarian tissue and the immunohistochemistry of follicle stimulating hormone receptor(FSHR)and luteinizing hormone receptor(LHR)after ASFV infection.Results showed that ASFV can infect GCs,the transcription level of inflammatory related factors in ASFV infected ovarian tissue was significantly increased compared with control group.Massive congestion,lymphocyte infiltration and vacuolization and degeneration of luteal cells in ovarian corpus luteum and stroma were found with microscopic observation.Macrophages infiltrated around necrotic cells in follicles.ASFV infection significantly decreased the transcription levels of FSHR and LHR.IHC results sindicated that ASFV infection significantly decreased the density of FSHR and LHR positive cells in ovary tissue.The results of this study provided references and accumulated useful datas for sow reproductive system pathological change and reproductive disorders caused by ASFV infection.
