摘要
目的:针对红花主产区变迁致其道地性特征淡化的现状,采用大鼠软组织损伤模型观察比较四川、新疆红花和云南白药的药效作用,为红花的道地性评价提供依据。方法:收集四川简阳和新疆昌吉产地红花,自制红花酊。用砝码击打法建立大鼠左后肢急性软组织损伤模型,分为模型对照组、简阳红花酊(JY)高、低(0.2 g/mL、0.1 g/mL)剂量组、昌吉红花酊(CJ)高、低(0.2 g/mL、0.1 g/mL)剂量组和云南白药酊组,另取正常大鼠为正常对照组。外用涂抹给药4 d,3次/d;取血检测各组大鼠血液流变学指标,酶联免疫法检测大鼠血清中白细胞介素-10(IL-10)和肿瘤坏死因子α(TNF-α)水平,并取损伤侧局部肌肉进行病理学检测。结果:与正常对照组比较,模型对照组大鼠外周血全血粘度(1、5、10、30、50、60 S^(-1))、红细胞聚集指数、卡松黏度均明显升高,差异有统计学意义(P<0.05);模型对照组大鼠血清TNF-α、IL-10水平均较正常对照组升高,差异有统计学意义(P<0.05);模型对照组大鼠病理组织学见肌组织广泛炎性细胞浸润。与模型对照组比较,昌吉红花酊0.2 g/mL、0.1 g/mL组可明显降低大鼠全血粘度(1、5、10、30、50、60 S^(-1))和红细胞聚集指数,且0.1 g/mL、0.2 g/mL组可分别降低红细胞聚集指数和卡松粘度值,差异有统计学意义(P<0.05);云南白药作用与昌吉红花作用相近;简阳红花对上述血液流变学指标也有降低作用趋势,但无统计学意义;新疆红花高、低剂量和云南白药均可明显降低大鼠血清TNF-α、IL-10水平,四川红花可降低血清IL-10水平,差异有统计学意义(P<0.05);各组大鼠损伤组织中炎性细胞浸润减轻。结论:四川红花和新疆红花酊均对大鼠软组织损伤有治疗作用,但在实验剂量下新疆红花作用更佳。
Objective:To observe and compare the pharmacodynamic effects of Carthamus tinctorius L.from Sichuan,Xinjiang and Yunnan Baiyao in order to provide a basis for the evaluation of Carthamus tinctorius L.from different producing areas on soft tissue injury in rats,aiming at the current situation that the geo-herbalism of Carthamus tinctorius L.were weakened due to the change of main producing areas of Carthamus tinctorius L.Methods:Carthamus tinctorius L.from Jianyang of Sichuan and Changji of Xinjiang were collected and tincture of Carthamus tinctorius L.was made by ourselves.The acute soft tissue injury model of left rear limb of rats was established by weight beating method.The rats were divided into model control group,Jianyang Carthamus tinctorius L.Tincture(JY)high-dose and low-dose(0.2 g/mL,0.1 g/mL)groups,Changji Carthamus tinctorius L.Tincture high-dose and low-dose(0.2 g/mL,0.1 g/mL)groups and Yunnan Baiyao Tincture group.Normal rats were also selected as normal control group.External daub for 4 days,3 times a day;Then blood samples were taken to detect the hemorheology of rats in each group.The levels of interleukin-10(IL-10)and tumor necrosis factorα(TNF-α)in serum of rats were determined by enzyme linked immunoassay.The local muscle of the injured side thigh was taken for pathological examination.Results:Compared with normal control group,the whole blood viscosity(1,5,10,30,50,60 S^(-1)),erythrocyte aggregation index and Casson viscosity of peripheral blood of rats in model control group were significantly increased,with statistical significance(P<0.05).The serum levels of TNF-αand IL-10 in model control group were higher than those in normal control group,and the difference was statistically significant(P<0.05).The histopathology of model control group showed extensive inflammatory cell infiltration in the muscle tissue.Compared with model control group,the whole bloodviscosity(1,5,10,30,50,60 S^(-1))and erythrocyte aggregation index of rats were significantly reduced in the 0.2 g/mL and 0.1 g/mL Changji Carthamus tinctorius L.groups,and the erythrocyte aggregation index and casson viscosity were decreased in the 0.1 g/mL and 0.2 g/mL groups,respectively.The difference was statistically significant(P<0.05);The effect of Yunnan Baiyao is similar to that of Changji Carthamus tinctorius L.Jianyang Carthamus tinctorius L.also had a decreasing effect on the above hemorheology indexes,but there was no statistical significance.High and low doses of Carthamus tinctorius L.from Xinjiang and Yunnan Baiyao could significantly reduce the levels of TNF-αand IL-10 in serum of rats,and Carthamus tinctorius L.from Sichuan could reduce the level of IL-10 in serum,the difference was statistically significant(P<0.05).The infiltration of inflammatory cells in the injured tissues was reduced in each group.Conclusion:Both Sichuan Carthamus tinctorius L.and Xinjiang Carthamus tinctorius L.tincture have therapeutic effects on soft tissue injury in rats,but the effect of Xinjiang Carthamus tinctorius L.is better at experimental dose.
