摘要
目的探讨自主研发的黑色二氧化钛(TiO_(2))介导的光动力学疗法对前列腺癌的杀伤作用。方法利用固相原位热还原法成功制备黑色TiO_(2),用不同浓度(0、31.25、62.50、125.00、250.00、500.00μg/mL)的黑色TiO_(2)(b-TP-700)处理LNCaP细胞12、24、48 h,利用CCK-8及荧光共聚焦显微镜来检测黑色TiO_(2)对前列腺癌细胞的毒性。利用不同浓度的黑色TiO_(2)(31.25、62.50、125.00、250.00、500.00μg/mL)处理LNCaP细胞后,利用激光(1 W/cm,808 nm)照射细胞0~7 min。利用CCK-8来检测黑色TiO_(2)介导的光动力学疗法对前列腺癌细胞的杀伤作用,利用荧光共聚焦显微镜检测黑色TiO_(2)介导的光动力疗法过程中产生活性氧的量。利用黑色TiO_(2)介导的光动力疗法处理LNCaP细胞后,通过裸鼠成瘤实验检测LNCaP细胞的裸鼠成瘤能力。结果不同浓度的黑色TiO_(2)作用于LNCaP细胞后,LNCaP细胞的活力及形态无明显变化(P>0.05)。在相同浓度的黑色TiO_(2)作用于LNCaP细胞时,随着激光激发时间的延长,LNCaP细胞的活力明显下降。另一方面,在激光激发时间相同的情况下,随着黑色TiO_(2)浓度的增加,LNCaP细胞的活力明显下降(P<0.05)。而且在相同浓度的黑色TiO_(2)作用下,激光照射组具有良好的ROS产生能力。在裸鼠成瘤实验,利用黑色TiO_(2)介导的光动力学疗法处理过的LNCaP细胞的成瘤能力明显降低(P<0.05)。结论新型光敏剂黑色TiO_(2)介导的光动力学疗法对前列腺癌细胞具有明显的杀伤作用,有望成为前列腺癌治疗的新方法。
Objective To investigate the killing effect of black titanium dioxide mediated photodynamic therapy on prostate cancer.Methods Black titanium dioxide was successfully prepared by solid-phase in-situ thermal reduc-tion.LNCaP cells were treated with different concentrations of black titanium dioxide(b-TP-700)(0,31.25,62.50,125.00,250.00,and 500.00μg/mL)for 12 h,24 h and 48 h.The toxicity of black titanium dioxide to prostate cancer cells was detected by CCK-8 and fluorescence confocal microscopy.LNCaP cells were treated with different concentra-tions of black titanium dioxide(31.25,62.50,125.00,250.00,and 500.00μg/mL)and irradiated with laser(1 W/cm,808 nm)for 0-7 minutes.CCK-8 was used to detect the killing effect of black titanium dioxide photodynamic ther-apy on prostate cancer cells,and fluorescence confocal microscopy was used to detect the amount of reactive oxygen spe-cies produced during black titanium dioxide photodynamic therapy.LNCaP cells were treated with photodynamic therapy mediated by black titanium dioxide,the tumorigenicity of LNCaP cells in nude mice was also detected.Results LNCaP cells had no significant change in activity and morphology after treated with different concentrations of black titanium diox-ide(P>0.05).When LNCaP cells were treated by the same concentration of black titanium dioxide,the activity of LN-CaP cells significantly decreased with the extension of laser excitation time.On the other hand,under the same laser exci-tation time,the viability of LNCaP cells significantly decreased with the increase of black titanium dioxide concentration(P<0.05).Moreover,under the same concentration of black titanium dioxide,the laser irradiation group had a good ROS production ability.In the tumorigenesis experiment of nude mice,we found that the tumorigenesis ability of LNCaP cells treated with black titanium dioxide mediated photodynamic therapy was significantly reduced(P<0.05).Conclu-sion Photodynamic therapy mediated by black titanium dioxide,a novel photosensitizer,has a significant killing effect on prostate cancer cells and is expected to be a new treatment method for prostate cancer.
