贵州地方水稻种质的SSR遗传多样性及群体结构分析 被引量:4

Genetic Diversity and Population Structure Analysis of Rice Landrace Germplasm from Guizhou Province by SSR Markers

摘要 为探明贵州地方水稻种质资源的遗传基础,本研究利用53对SSR引物对来源于贵州省内多个地区的273份地方水稻种质进行遗传多样性和群体结构分析。研究结果表明,53个SSR标记共检测到182个等位基因,每个位点的等位基因数变化范围为2〜6,平均为3.43;平均有效等位基因数为2.0145;Shannon信息指数变化范围为0.2620-1.5828,平均为0.7788;Nei's基因多样性指数变化范围为0.1333-0.7723,平均为0.4325;多态信息含量变化范围为0.1266-0.7366,平均为0.3680。基于Nei's遗传距离的UPGMA聚类分析将273份资源划分为2个类群,分别包含240和33份材料;Structure群体结构分析结果划分为2个类群,分别包含242和31份材料;其中262份(95.97%)材料的Q值≥0.9,仅2份材料Q<0.6,划分到混合群。群体结构分析结果与UPGMA聚类结果大致相同。综上所述,贵州地方水稻种质资源的遗传多样性不够丰富,遗传结构基础较为单一,造成这一现象的原因可能是贵州地方水稻种质由相同或类似的种质在各地区进行流转发展而来。本研究结果为贵州地方水稻资源的有效保护和高效利用提供科学依据。 In order to ascertain the genetic relationship of rice landrace germplasm resources from Guizhou Province, 53 pairs of SSR primers were used to analyze the genetic diversity and population structure of 273 rice landrace germplasms from several areas in Guizhou Province. A total of 182 alleles were detected from 53 SSR markers, and the number of alleles per locus ranged from 2 to 6, with an average of 3.43. The average effective number of alleles was 2.014 5. Shannon’s information index ranged from 0.262 0 to 1.582 8, with an average of 0.778 8. Nei’s gene diversity index ranged from 0.133 3 to 0.772 3, with an average of 0.432 5. The polymorphism information content varied from 0.126 6 to 0.736 6, with an average of 0.368 0. UPGMA clustering analysis based on Nei’s genetic distance divided 273 resources into two groups, including 240 and 33 materials, respectively. The results of population structure analysis were divided into two groups, including 242 and 31 materials, respectively.Among 273 rice landrace germplasms, Q values of 262 materials(95.97%) were greater than or equal to 0.9, and only 2 materials were less than 0.6 in Q value, which were classified into admixed group. The results of population structure analysis were similar to those of UPGMA clustering. In conclusion, the genetic diversity of rice landrace germplasm resources from Guizhou Province is not rich enough, and the basis of genetic structure is relatively simple, which may due to that Guizhou rice landrace germplasms were developed from the circulation of the same or similar germplasms in different regions. These results have provided scientific basis for effective protection and efficient utilization of rice resources.
