基于HS-SPME-GC-MS的小罗伞根挥发性成分分析条件优化

Optimization of Extraction Conditions of Volatile Components for Ardisia punctata Lindl.Roots Based on HS-SPME-GC-MS

摘要 建立基于HS-SPME-GC-MS法的快速测定小罗伞根挥发性成分的方法。以总峰面积为考察指标,在单因素(萃取头极性、萃取温度、萃取时间、平衡时间、解吸时间等)实验的基础上,通过L_(9)(3^(4))四因素三水平正交实验进行优化。结果显示:最佳条件为萃取针CAR/DVB/PDMS,萃取温度80℃、平衡时间20min、萃取时间50 min、解吸时间4 min;在此条件下,从罗浮山小罗伞根部鉴定出27种挥发性成分,主要为α-古巴烯(27.59%)、水杨酸甲酯(9.04%)、α-姜黄烯(8.58%)、α-雪松烯(8.46%)。本研究结果可为快速分析小罗伞根的挥发性成分提供科学依据。 To establish a rapid method for the determination of volatile components of Ardisia punctata Lindl.roots based on HS-SPME-GC-MS.Based on the single factor(polarity of extraction fiber,extraction temperature,extraction time,balance time,desorption time,etc.)experiments,the total peak area was carried out as index,an L_(9)(3^(4))orthogonal experiments were carried out to optimize the extraction conditions.The optimum conditions were as follows:CAR/DVB/PDMS extraction head,temperature of 80℃,equilibrium time of 20 min,extraction time of 50 min and desorption time of 4 min.Under these conditions,27 volatile components were identified from Ardisia punctata Lindl.roots,they were mainlyα-cubanene(27.59%),methyl salicylate(9.04%),α-curcumene(8.58%)andα-cedarene(8.46%).The results can provide scientific basis for rapid analysis of the volatile components of Ardisia punctata Lindl.roots.
