头腹针结合颈通康对颈动脉狭窄脑血流低灌注模型大鼠干预机制研究 被引量:3

Intervention Mechanism of Scalp and Abdominal Acupuncture Combined with Jing Tong Kang in Rat Model of CAS with Cerebral Blood Flow Hypoperfusion

摘要 目的:分析头腹针疗法结合颈通康对颈动脉狭窄脑血流低灌注模型大鼠的作用机制。方法:健康雄性Wistar大鼠50只,采用“针控线栓法”进行颈动脉狭窄造模,成功后随机分为模型组、中药治疗组和针药治疗组,每组各15只模型大鼠,假手术组5只,不操作。治疗前、7 d、14 d和21 d共4个时间点采用神经功能评分、行为学评估;3 d、7 d和14 d原位杂交检测NCAM、免疫荧光标记及激光共聚焦检测GFAP、NeuN。结果:神经功能评分、行为学评估7 d、14 d评分治疗组优于模型组,针药组优于中药组(P<0.05或P<0.01)。NCAM、GFAP和NeuN的阳性表达均在7 d达到高峰,在3 d、7 d和14 d治疗组均优于模型组,针药组优于中药组(P<0.05或P<0.01)。结论:头腹针疗法结合颈通康对颈动脉狭窄脑血流低灌注模型大鼠神经血管单元有干预作用。 Objective: To analyze the intervention mechanism of scalp and abdominal acupuncture combined with Jing Tong Kang in rat model of carotid artery stenosis(CAS) with cerebral blood flow hypoperfusion. Methods: 50 healthy male Wistar rats were randomly divided into the model group(n=15), the TCM medication group(n=15), the acupuncture and TCM medication group(n=15) and the sham-operation group(n=5). The carotid stenosis model was established by ‘needle controlled thread bolt method’. Neurological function score was observed and behavioral assessment was carried out before the treatment, after 7 days, 14 days and 21 days of the treatment. Immunofluorescence labeling and laser confocal detection were applied to detect GFAP and NeuN. In situ hybridization(ISH) was used to detect NCAM after 3 days, 7 days and 14 days of the treatment. Results: The scores of nerve function and behavior assessment were better in the treatment groups than those in the model group after 7 days and 14 days of the treatment, and the acupuncture and TCM medication group was better than the TCM medication group(P<0.05, P<0.01). The positive expressions of NCAM, GFAP and NeuN reached peak after 7 days of the treatment;the treatment groups were superior to the model group, and the acupuncture group and TCM medication group was superior to the TCM medication group after 3 days, 7 days and 14 days of the treatment(P<0.05, P<0.01). Conclusion: Scalp and abdominal acupuncture combined with Jing Tong Kang has an intervention effect on neurovascular unit of CAS rat model with cerebral blood flow hypoperfusion.
