辛温通阳中药葱白提取物对高脂血症模型大鼠线粒体代谢相关限速酶表达的影响 被引量:4

摘要 目的:探讨辛温通阳中药葱白提取物对高脂血症模型大鼠线粒体代谢相关限速酶表达的影响。方法:30只健康雄性SD大鼠随机分为对照组、模型组、葱白低剂量组、葱白中剂量组、葱白高剂量组与辛伐他汀组,除对照组外,其他组均通过脂肪乳灌胃法构建高脂血症模型。对照组与模型组每天灌胃生理盐水,葱白低、中、高剂量组分别按照25 mg/kg、50 mg/kg、70 mg/kg灌胃葱白提取物,辛伐他汀组灌胃100 mg/kg辛伐他汀;造模成功后采用全自动生化分析仪检测血清中TC、TG、HDL-C及LDL-C水平;运用RT-PCR检测肝脏细胞内己糖激酶(HK1)、环氧合酶2(COX-2)、肉毒碱棕榈酰转移酶1(CPTl)、柠檬酸合酶(CS)、细胞色素C(Cyc-C)mRNA表达水平;运用Western-blot法检测肝脏细胞内HK1、COX-2、CPTl、CS与Cyc-C蛋白表达水平。结果:连续给药4周后,与模型组相比,葱白高剂量组的TC、TG、LDL-C水平显著降低(P<0.05),而HDL-C水平则显著上升(P<0.05);RT-PCR结果显示,给药4周后,与模型组相比,葱白高剂量组COX-2基因表达水平显著降低(P<0.05),而HK1、CS、CPT1及Cyc-C的mRNA表达水平显著升高(P<0.05);Western blot结果显示,与模型组相比,葱白高剂量组COX-2蛋白表达水平显著降低(P<0.05),而HK1、CS、CPT1及Cyc-C的蛋白表达水平显著升高(P<0.05)。结论:中药葱白提取物对高脂血症模型大鼠有明显的降血脂作用,能通过调节线粒体代谢相关限速酶基因及蛋白的表达来促进脂质代谢。 Objective:To investigate the effect of fistular onion stalk extract on the gene expression of rate limiting enzymes associated mitochondrial metabolism in hyperlipidemia(HLP)model rats.Methods:30 male SD rats were randomly divided into the control group,the model group,the fistular onion stalk extract groups of low-dose,medium-dose and high-dose,as well as the Simvastatin group.Rats in all groups except the control group were gavaged with fat milk to construct HLP model.The control group and the model group were gavaged with saline everyday.The fistular onion stalk extract groups were gavaged with fistular onion stalk extract at the doses of 25 mg/kg,50 mg/kg and 70 mg/kg,respectively.The Simvastatin group was gavaged with Simvastatin at the dose of 100 mg/kg.After establishing the HLP model,the serum levels of TC,TG,HDL-C and LDL-C were detected by biochemical examination.RT-PCR was applied to detect the mRNA expressions of HK1,COX-2,CPTl,CS and Cyc-C in liver cells.Western-blot was used to detect the protein expressions of HK1,COX-2,CPTl,CS and Cyc-C in liver cells.Results:After four-week continuous administration,the serum levels of TC,TG and LDL-C were decreased,and the serum level of HLD-C was increased in the high-dose group of fistular onion stalk extract compared with those in the model group(P<0.05).RT-PCR results showed that the mRNA expression of COX-2 was down-regulated,and the mRNA expressions of HK1,CPTl,CS and Cyc-C were up-regulated in the high-dose group compared with those in the model group(P<0.05).The mRNA expression of COX-2 was significantly reduced,and the mRNA expressions of HK1,CPTl,CS and Cyc-C were significantly increased in the high-dose group compared with those in the model group(P<0.05).Western blot results showed that the protein expression of COX-2 was decreased,and the protein expressions of HK1,CS,CPT1 and Cyc-C were increased in the high-dose group compared with those in the model group.Conclusion:Fistular onion stalk extract showed obvious lipid-reducing effects in hyperlipidemic rats,which promotes lipid metabolism by regulating the gene expression of rate limiting enzymes associated mitochondrial metabolism.
