摘要
目的探究罗哌卡因对宫颈癌细胞C-33A生长、侵袭和上皮间质转化的影响及裸鼠移植瘤生长的影响。方法将细胞随机分为4组:罗哌卡因0.00 mmol/L组、罗哌卡因0.25 mmol/L组、罗哌卡因0.50 mmol/L组和罗哌卡因1.00 mmol/L组。5-乙炔基-2′脱氧尿嘧啶核苷(EDU)染色检测细胞增殖;Transwell检测细胞侵袭;显微镜观察上皮间质转化形态学变化;蛋白免疫印迹检测波形蛋白(Vimentin)、纤维黏连蛋白(Fibronectin)蛋白表达水平。裸鼠皮下注射宫颈癌细胞C-33A将裸鼠随机分为2组:罗哌卡因0.0 mg/kg组和罗哌卡因0.3 mg/kg组,检测裸鼠肿瘤重量,脱氧核糖核苷酸末端转移酶介导的缺口末端标记(TUNEL)染色检测裸鼠肿瘤细胞凋亡,免疫组化检测Vimentin阳性细胞数。结果细胞实验中,与罗哌卡因0.00 mmol/L组相比较,罗哌卡因0.50、1.00 mmol/L组EDU阳性细胞数显著降低,侵袭细胞数显著降低,上皮间质转化受到抑制,Vimentin、Fibronectin蛋白水平显著降低(均P<0.05);裸鼠实验中,与罗哌卡因0.0 mg/kg组相比较,罗哌卡因0.3 mg/kg组第25、30天肿瘤体积显著变小,肿瘤重量显著降低,细胞凋亡率显著升高,Vimentin阳性细胞数显著降低(P<0.05)。结论罗哌卡因抑制宫颈癌细胞C-33A增殖、侵袭、上皮间质转化和裸鼠移植瘤肿瘤的形成。
Objective To investigate the effects of ropivacaine on the growth,invasion and interstitial transformation of cervical cancer cells C-33A and the effect on xenograft tumor growth in nude mice.Methods The cells were randomly divided into Ropivacaine 0.00,0.25,0.50 and 1.00 mmol/L groups.Cell proliferation was detected by 5-ethynyl-2'-deoxyuridine(EDU)staining.Transwell was used to detect cell invasion;morphological changes of epithelial mesenchymal transformation were observed microscopically.The expression levels of Vimentin and Fibronectin protein were detected by Western blotting.Nude mice were randomly divided into 2 groups:Ropivacaine 0.0 and 0.3 mg/kg groups by subcutaneous injection of cervical cancer cell C-33A.Tumor weight was detected in nude mice.Tumor cell apoptosis was detected by TUNEL staining.Immunohistochemical method was used to detect Vimentin positive cell number.Results Compared with Ropivacaine 0.0 mmol/L group,the number of EDU positive cells in Ropivacaine 0.5 and 1.0 mmol/L groups was significantly reduced(P<0.05),the number of invasive cells was significantly reduced(P<0.05),the epithelial mesenchymal transformation was obviously inhibited,and the levels of Vimentin and Fibronectin proteins were significantly reduced(P<0.05).In nude mice,compared with Ropivacaine 0.0 mg/kg group,the tumor weight of Ropivacaine 0.3 mg/kg group was significantly reduced(P<0.05),tumor volume was significantly reduced on day 30(P<0.05),the apoptosis rate was significantly increased(P<0.05),and the number of Vimentin positive cells was significantly reduced(P<0.05).Conclusions Ropivacaine could inhibit C-33A proliferation,invasion,epithelial mesenchymal transformation and tumor formation in nude mice.
