摘要
综合性实验在培养学生创新思维及新药研发能力中发挥着重要作用,因而开展了从较为成熟的药物合成科研实验“氨基甾体类衍生物的合成”转化成有机化学综合教学实验的探索。以雄烯二酮为原料,通过羰基的保护、选择性脱保护、羟醛缩合及环加成反应合成了甾体D环并吡唑啉衍生物,并运用核磁共振技术^(1)H NMR,^(13)C NMR和1DNOE研究了其结构和构型。教学实践表明,该实验巩固了有机化学的理论知识,开拓了学生的视野,培养了学生有机合成实验操作和谱图解析等方面的综合能力,开发了学生的创新潜能,为培养创新型人才奠定了基础。
The design experiment plays an important role in cultivating students’innovative thinking and new drug research and development capabilities.This work provides a researchoriented comprehensive organic chemistry teaching experiment converted from a scientific research achievement in the synthesis of aminosteroidal derivatives.Steroidal D-ring fused pyrazoline derivative is synthesized from androstenedione via carbonyl protection,selective deprotection,aldol condensation and cycloaddition reaction.Its structure and configuration are identified by ^(1)H NMR,^(13)C NMR and 1D NOE.The teaching practice shows that the experiment consolidates the theoretical knowledge of organic chemistry,broadens students’vision,cultivates their comprehensive research ability in synthetic experiment operation and spectrum analysis,develops their innovation potential,and lays a foundation for cultivating innovative talents.
