氟对成骨细胞骨钙素和Ⅰ型胶原基因表达的影响 被引量:2

Effects of fluoride on gene expression of osteocalcin and typeⅠcollagen in osteoblasts

摘要 目的探讨氟对成骨细胞骨钙素基因(OCN)和Ⅰ型胶原蛋白基因(COL1)表达的影响。方法采用人成骨肉瘤细胞系(Saos-2)建立体外染氟模型,即经体外细胞培养后分别以不同浓度梯度氟化钠(0.0 mg/L,5.0 mg/L,10.0 mg/L,20.0 mg/L和40.0 mg/L NaF)处理;在实验组中加入坏死凋亡抑制剂(Necrostatin-1,Nec-1)抑制细胞的程序性坏死,提取细胞的总mRNA,通过实时荧光定量RT-PCR方法,分析染氟对Saos-2细胞骨钙素基因和Ⅰ型胶原蛋白基因表达的影响。结果与对照组相比,经5.0 mg/L,10.0 mg/L,20.0 mg/L和40.0 mg/L NaF处理后,OCN和COL1的表达域上调,其中以(5.0~10.0 mg/L)增强的最明显。低浓度氟化钠(5.0~10.0 mg/L)可以促进OCN和COL1的基因表达,高浓度(>20.0 mg/L)氟化钠对OCN和COL1基因的相对表达量上调程度较低;加入Nec-1的组OCN和COL1基因表达域较对照组上调明显。结论在基因水平上,低浓度氟化钠(5.0~10.0 mg/L)上调人成骨细胞中OCN和COL1基因的表达;高浓度氟化钠(>20.0 mg/L)对OCN、COL1基因的相对表达量的上调程度较低。Nec-1对两基因的表达均为上调。氟诱导的程序性坏死能够影响成骨相关基因OCN和COL1的表达。 Objective To investigate the influence of fluoride on the expression of osteocalcin gene(OCN)and typeⅠcollagen gene(COL1)in osteoblasts.Methods The human osteosarcoma cell line(Saos-2)was used to establish the fluoride exposure model in vitro that was the cells were respectively treated with different concentration gradients of sodium fluoride(NaF,0.0,5.0,10.0,20.0 and 40.0 mg/L)after cell culture in vitro.An inhibitor of necrosis and apoptosis(Necrostatin-1,Nec-1)was added in the experimental group for inhibiting the programmed necrosis of the cells.The total cell mRNA were extracted and detected with real-time fluorescence quantitative PCR for analyzing the influence of fluoride exposure to the expression of osteocalcin gene and type I collagen gene in Saos-2 cells.Results Compared with the control group,the expression domains of OCN and COL1 were up-regulated after NaF exposure of 5.0 mg/L,10.0 mg/L,20.0 mg/L and40.0 mg/L,especially enhanced in the concentration of 5.0-10.0 mg/L.Low concentration of NaF(5.0-10.0 mg/L)could promote the gene expression of OCN and COL1,and the high concentration of NaF(>20.0 mg/L)only enhanced the expression domains at low degree.The results showed the expression domains of OCN and COL1 genes obviously up-regulated higher in the group of adding Nec-1 than in the control group.Conclusions Low concentration of NaF(5.0-10.0 mg/L)can upregulate the genes expression of OCN and COL1 in human osteoblasts at the gene level,but high concentration of NaF(>20.0 mg/L)enhanced the expression domains at low degree.Nec-1 can up-regulate the expression of both genes.The expression of osteogenesis-related genes of OCN and COL1 can be influenced by the procedural necrosis induced by fluoride.
