摘要
目的 通过液相色谱-质谱串联技术研究槐耳清膏治疗阿霉素肾病大鼠的作用机制。方法 将9只6周龄清洁级Wistar大鼠随机分为对照组、阿霉素肾病模型组及槐耳清膏治疗组(n=3),通过液相色谱-质谱串联技术对造模4周的大鼠尿标本进行分析,筛选显著差异表达的尿蛋白质,对差异蛋白进行功能富集分析。结果 (1)3组大鼠相互比较,鉴定出285种可定量分析的差异蛋白质;(2)差异蛋白显著富集的生物过程包括急性炎症反应、成纤维细胞增殖、凝血级联反应、免疫激活等;(3)肾小球硬化时细胞外基质(extracellular matrix, ECM)发生变化的2种蛋白质在尿中显著差异表达,包括α-1抗蛋白酶、胶原α-1(Ⅰ)链。结论 局灶节段性肾小球硬化(focal segmental glomerulosclerosis, FSGS)的发病可能与急性炎症反应、ECM失调有关,槐耳清膏治疗FSGS的机制可能与降低凝血级联反应、抑制免疫活化和炎症反应密切相关。
Objective To study the mechanism of Huaier aqueous extract in the treatment of adriamycin nephropathy rats by liquid chromatography-mass spectrometry(LC-MS). Methods Nine clean grade Wistar rats aged 6 weeks were randomly divided into control group, adriamycin nephropathy model group and Huaier aqueous extract treatment group(n=3). The urine samples of rats after 4 weeks of modeling were analyzed by liquid chromatography-mass spectrometry(LC-MS), and the urine proteins with significant difference in expression were screened, and the functional enrichment analysis of the differential proteins was performed. Results(1)Three groups of rats were compared with each other, 285 kinds of differential proteins that can be analyzed quantitatively were identifed.(2)The biological processes of significant enrichment of differential proteins included acute inflammatory reaction, fibroblast proliferation, coagulation cascade reaction, and immune activation.(3)Two proteins related to glomerular ECM were screened out in the urine protein with significant differences, including alpha-1-antiproteinase and collagen alpha-1(Ⅰ) chain. Conclusion The pathogenesis of focal segmental glomerulosclerosis(FSGS) may be related to acute inflammatory response and ECM disorder. The mechanism of Huaier aqueous extract in the treatment of FSGS may be closely related to reducing coagulation cascade reaction, inhibiting immune activation and inflammatory response.
