陶湖杂交F1代与萨湖杂交F1代羔羊背最长肌转录组分析 被引量:3

Transcriptome analysis of longissimus dorsi muscle of Taohu F1 and Sahu F1 lambs

摘要 试验旨在对陶湖杂交F1代与萨湖杂交F1代羔羊背最长肌进行转录组测序分析,筛选肉质相关基因。选择饲养条件相同、健康无病的陶塞特与湖羊杂交F1代羔羊(TH)和萨福克与湖羊杂交F1代羔羊(SH)各3只屠宰,采集背最长肌,利用RNA-Seq技术测序,筛选差异表达基因,对其进行GO和KEGG富集分析。结果显示:筛选得到552个差异表达基因,其中385个上调基因,167个下调基因。GO富集显示,差异表达基因主要富集在免疫反应、蛋白质折叠、Ras蛋白信号转导、MHC蛋白复合物、质膜蛋白复合物、肌动蛋白细胞骨架、未折叠蛋白结合、细胞外基质结构成分等条目上。KEGG富集显示,与肉质相关的通路主要为PPAR信号通路、碳水化合物消化吸收通路、AMPK信号通路、脂类代谢通路等,筛选到与肉质相关的基因有SCD、LIPG、LPL。研究表明,SCD、LIPG、LPL基因在羔羊的肉质形成过程中发挥了重要作用,可作为肉质性状的候选基因。 The study was to perform transcriptome sequencing analysis of longissimus dorsi of Taohu hybrid F1generation and Sahu hybrid F1 generation lambs, to screen meat quality-related genes. Three healthy and disease free Tausset and Hu sheep F1 lambs(TH) and Suffolk and Hu sheep F1 lambs(SH) were slaughtered, and their longissimus dorsi muscle were analyzed by transcriptome sequencing, the differentially expressed genes were analyzed by GO and KEGG enrichment. The results showed that a total of 552 differentially expressed genes were screened, including 385 upregulated genes and 167 down-regulated genes. GO enrichment showed that the differentially expressed genes were mainly enriched in antigen processing and presentation, immune response, protein folding, Ras protein signal transduction,MHC protein complex, plasma membrane protein complex, myosin complex, actin cytoskeleton, unfolded protein binding,extracellular matrix structural components and other components. KEGG enrichment showed that the main pathways related to meat quality were PPAR signaling pathway, carbohydrate digestion and absorption, AMPK signaling pathway and lipid metabolism pathway, etc. SCD, LIPG and LPL genes were screened for meat quality. The study indicates that SCD, LIPG and LPL genes plays an important role in the process of meat quality formation of lambs, which can be used as candidate genes for meat quality traits.
