樱桃李多酚类物质成分鉴定及其改善脂代谢的作用研究 被引量:3

摘要 目的:分析樱桃李果实中多酚类物质的成分及其对肥胖小鼠脂代谢的改善作用,为开发降脂功能食品提供一定的理论基础。方法:以樱桃李多酚提取物为材料,采用液相色谱-质谱分析其主要的多酚类物质,进一步建立肥胖小鼠模型,采用不同剂量多酚灌胃处理,8周后处死小鼠,采集脏器和血清,计算脏器指数,检测血清中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)水平;HE和油红O染色对肝脏进行组织形态学分析;荧光定量PCR和Western blot分别检测脂代谢相关因子的mRNA和蛋白表达。结果:(1)多酚提取物中鉴定出18种多酚单体,包括5种酚酸和单宁,13种黄酮及其衍生物。(2)樱桃李多酚处理可显著降低肥胖小鼠体重,并降低血清AST和ALT活性及TG、TC和LDL-C含量,升高HDL-C含量,保护小鼠器官,抑制肝脏脂质积累。(3)樱桃李多酚显著下调过氧化物酶体增殖物激活受体γ(PPARγ)、脂肪酸合酶(FAS)、乙酰辅酶A羧化酶(ACC)、CD36、胆固醇调节元件结合蛋白1c(SREBP-1c)和CCAAT/增强子结合蛋白α(C/EBPα)的mRNA表达,上调肉碱棕桐酰转移酶1(CPT-1)和酰基辅酶A氧化酶(ACO)的mRNA表达。(4)樱桃李多酚显著下调FAS、SREBP-1c、C/EBPα和CD36蛋白表达,上调磷酸化AMP活化蛋白激酶(p-AMPK)与AMPK比值。结论:樱桃李多酚可通过激活AMPK信号通路改善肥胖小鼠脂代谢。 AIM:To explore the effect of polyphenols from Prunus cerasifera fruit on lipid metabolism in obese mice.METHODS:Polyphenols extracted from Prunus cerasifera Ehr. were analyzed by liquid chromotography with mass spectrometry. The mice were intragastrically administered with different doses of the polyphenols for 8 weeks. After the mice were sacrificed,their organs were collected to calculate the organ indexes. The levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)in serum were measured. The morphological changes of liver tissues were observed by HE and oil red O staining. The expression of lipid metabolism-related molecules at mRNA and protein levels was detected by fluorescence quantitative PCR and Western blot,respectively.RESULTS:(1)Totally 18polyphenolic monomers were identified in the polyphenol extract,including 5 phenolic acids and tannins,and 13 flavonoids and their derivatives.(2)Treatment with Prunus cerasifera polyphenols significantly decreased the body weight,reduced serum AST,ALT,TG,TC and LDL-C levels,increased HDL-C content,and inhibited liver lipid accumulation in obese mice.(3)Prunus cerasifera polyphenols significantly down-regulated the mRNA expression of peroxisome roliferator-activated receptor γ(PPARγ),fatty acid synthase(FAS),acetyl-CoA carboxylase(ACC),CD36,sterol regulatory element-binding protein-1c(SREBP-1c)and CCAAT/enhancer-binding protein α(C/EBPα),but up-regulated the mRNA expression of carnitine palmitoyltransferase-1(CPT-1)and acyl-CoA oxidase(ACO).(4)Prunus cerasifera polyphenols significantly decreased the expression of FAS,SREBP-1c,C/EBPα and CD36 proteins,but increased the ratio of phosphorylated AMP-activated protein kinase(p-AMPK)to AMPK.CONCLUSION:Prunus cerasiferapolyphenols improve lipid metabolism in obese mice by activating AMPK signaling pathway.
