摘要
目的:瑞舒伐他汀(RSV)调节微小RNA(miR)-21表达对慢性牙周炎大鼠炎症反应的影响。方法:通过丝线结扎并固定的方法复制慢性牙周炎大鼠模型。将造模后的50只大鼠随机分为模型组、RSV组、antagomir NC(抑制剂阴性对照)组、miR-21抑制剂(miR-21 antagomir)组、RSV+miR-21 antagomir组,另取10只大鼠正常饮水喂食并以生理盐水干预,作为对照组。经药物、miR-21抑制剂分组干预后,检测牙龈指标-附着丧失(AL)和牙龈指数(GI);收集血清样本,检测各组大鼠血清中炎症因子指标白细胞介素-1β(IL-1β)、白细胞介素-17(IL-17)、肿瘤坏死因子-α(TNF-α)水平;qRT-PCR法检测血清中miR-21表达情况;分离牙周组织,检测各组大鼠病理学变化及基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-9(MMP-9)蛋白表达。结果:模型组牙周组织炎性浸润、水肿等病理状况较为严重,miR-21表达、AL、GI、IL-1β、IL-17、TNF-α水平、MMP-1、MMP-9蛋白表达均较对照组增加(P<0.05);与模型组相比,RSV组大鼠牙周组织病理状况逐渐缓解,AL、GI、IL-1β、IL-17、TNF-α水平、MMP-1、MMP-9蛋白表达较模型组均显著降低,miR-21表达增加(P<0.05);antagomir NC组各指标与模型组相比差异无统计学意义(P>0.05);与antagomir NC组相比,miR-21 antagomir组大鼠病理状况加重,AL、GI、IL-1β、IL-17、TNF-α水平、MMP-1、MMP-9蛋白表达均显著增加,miR-21表达降低(P<0.05);RSV+miR-21 antagomir组大鼠病理状况较RSV组明显加重,AL、GI、IL-1β、IL-17、TNF-α水平、MMP-1、MMP-9蛋白表达均显著增加,miR-21表达降低(P<0.05)。结论:RSV可以降低炎症反应,改善慢性牙周炎大鼠牙周组织病理状况以及牙周指标,可能与提高miR-21表达有关。
Objective:To explore the effects of rosuvastatin(RSV)regulating microRNA(miR)-21 expression on inflammatory response in rats with chronic periodontitis.Methods:The chronic periodontitis rat model was replicated by silk ligation and fixation.Totally 50 rats after modeling were randomly divided into model group,RSV group,antagomir NC(inhibitor negative control)group,miR-21 inhibitor(miR-21 antagomir)group,and RSV+miR-21 antagomir group.Another 10 rats were fed with normal drinking and diet,and intervened with normal saline as the control group.After intervention with drugs and miR-21 inhibitors,the gingival index-attachment loss(AL)and gingival index(GI)were detected;serum samples were collected to measure the levels of inflammatory factor indicators-interleukin-1β(IL-1β),IL-17 and tumor necrosis factor-α(TNF-α)in serum of rats in each group;the expression of miR-21 in serum was measured by qRT-PCR;the periodontal tissues were withdrawn,and the pathological changes and the protein expression of matrix metalloproteinase-1(MMP-1)and MMP-9 were measured in each group.Results:The pathological conditions of periodontal tissue,such as inflammatory infiltration and edema,were more serious in the model group,the expression of miR-21,levels of AL,GI,IL-1β,IL-17 and TNF-α,and the protein expression of MMP-1 and MMP-9 were all higher than those in the control group(P<0.05);compared with those in the model group,the pathological conditions of periodontal tissue in the RSV group were gradually relieved,and the levels of AL,GI,IL-1β,IL-17 and TNF-α,and the protein expression of MMP-1 and MMP-9 were obviously lower than those in the model group,and the miR-21 expression was increased(P<0.05);there was no obvious difference in each index between the antagomir NC group and the model group(P>0.05);compared with those in the antagomir NC group,the pathological conditions of rats in the miR-21 antagomir group were aggravated,and the levels of AL,GI,IL-1β,IL-17 and TNF-α,and the protein expression of MMP-1 and MMP-9 were obviously higher,and the miR-21 expression was decreased(P<0.05);compared with those in the RSV group,the pathological conditions of rats in the RSV+miR-21 antagomir group were obviously increased,and the levels of AL,GI,IL-1β,IL-17 and TNF-α,and the protein expression of MMP-1 and MMP-9 were obviously higher,and the miR-21 expression was decreased(P<0.05).Conclusion:RSV can reduce the inflammatory response and improve the pathological conditions of periodontal tissue and periodontal indexes in rats with chronic periodontitis,which may be related to the increase of miR-21 expression.
