HPLC法同时测定莱阳梨止咳颗粒、糖浆中的盐酸麻黄碱、绿原酸、苯甲酸的含量

摘要 目的建立高效液相色谱法测定莱阳梨止咳颗粒、糖浆中盐酸麻黄碱、绿原酸、苯甲酸的含量。方法Agilent C_(18)色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈-0.4%磷酸,梯度洗脱;检测波长:选用210 nm(盐酸麻黄碱)、327 nm(绿原酸)、230 nm(苯甲酸);柱温:30℃;流速:1 mL·min^(-1),进样量10μL。结果盐酸麻黄碱、绿原酸、苯甲酸的含量浓度分别在0.0622~0.6223、0.0116~0.1159、0.2123~5.3075(莱阳梨止咳颗粒)、1.0615~6.3690(莱阳梨止咳口服液)μg·mL^(-1)范围内与峰面积呈良好的线性关系(r=0.9999、1.0000、1.0000、0.9999);莱阳梨止咳颗粒平均加样回收率分别为97.85%、95.32%、96.62%(n=6);莱阳梨止咳糖浆平均加样回收率分别为94.19%、95.24%、97.74%(n=6)。结论所建立的测定方法简便、准确、重复性好、专属性强,可同时测定莱阳梨止咳颗粒、糖浆中的3种成分的含量。 Objective To develop an HPLC method for simultaneously determination of ephedrine hydrochloride,chlorogenic acid and benzoic acid in Laiyangli Cough Granules,Syrup.Methods Agilent C_(18)(4.6 mm×250 mm,5μm)column was adopted.The mobile phase was acetonitrile-0.4% phosphate with gradient elution at a flow rate of 1.0 mL·min^(-1).The column temperature was 30℃.The detection wavelength was set as 210 nm(ephedrine hydrochloride),327 nm(chlorogenic acid),230 nm(benzoic acid).Results The linear ranges of ephedrine hydrochloride,chlorogenic acid,benzoic acid were between 0.06223~0.6223μg·m L^(-1)(r=0.9999),0.01159~0.1159μg·mL(r=1.0000),0.2123~5.3075μg·mL^(-1)(Laiyangli Cough Granules)(r=1.0000),1.0615~6.3690μg·mL^(-1)(Laiyangli Cough Syrup)(r=1.0000).The average recoveries(n=6)were 97.89%,96.37%,96.63%,103.3%,respectively.Conclusion The method was simple,accurate and reproducible,and it can be used for quality control of Laiyangli Cough Granules,Syrup.
