摘要
香菇多糖是一种天然的活性物质,其制备方法常包含浓缩及醇沉操作。为提高香菇多糖的得率,采用单因素结合正交设计优化香菇多糖的浓缩及醇沉工艺。结果显示:浓缩比(原料质量:浓缩液体积)为1∶2g/mL时,多糖得率最高,由正交优化的最佳醇沉条件为:醇沉时间4h、乙醇用量4.5倍体积、乙醇浓度95%,此条件下所得多糖质量最大,影响醇沉效果的因素强弱为:乙醇浓度>醇沉时间>乙醇用量体积倍数。
Lentinan is a natural active substance,and its preparation methods usually includes concentration and alcohol precipitation.In order to improve the yield of lentinan,single factor combined with orthogonal design was used to optimize the concentration and alcohol precipitation process of lentinan.The results showed that when the concentration ratio(raw material quality:volume of concentrated liquid)was 1∶2g/mL,the yield of polysaccharide was the highest.The optimum ethanol precipitation conditions were as follows:ethanol precipitation time 4 h,ethanol dosage 4.5 times of volume,ethanol concentration 95%.Under these conditions,the quality of the polysaccharide obtained was the largest.The factors affecting the alcohol precipitation effect were as follows:ethanol concentration>ethanol precipitation time>volume multiple of ethanol dosage.
