摘要
目的 探究紫铆因对糖尿病大鼠肝脏胰岛素抵抗的改善作用及可能机制。方法 SD大鼠随机分为对照组(Control组)、糖尿病组(DM组)、紫铆因低剂量组(Butein-L组)和紫铆因高剂量组(Butein-H组),每组10只。测量大鼠空腹血糖和口服糖耐量;ELISA检测大鼠血清胰岛素、总胆固醇和甘油三酯水平以及肝组织TNF-α和IL-6水平;检测肝糖原含量;计算大鼠胰岛素抵抗指数和胰岛素敏感指数;HE染色观察肝组织病理学变化;Western blot检测肝组织IRS-1和GLUT4蛋白表达;免疫荧光检测肝组织SIRT1和NF-κB蛋白表达。结果 紫铆因降低糖尿病大鼠空腹血糖、血清胰岛素和血脂水平,增加肝糖原含量,降低胰岛素抵抗指数,增加胰岛素敏感指数,改善肝组织病理损伤,降低肝组织炎症因子水平,增加肝组织IRS-1、GLUT4和SIRT1蛋白表达水平,降低肝组织NF-κB蛋白表达水平。结论 紫铆因改善糖尿病大鼠肝组织胰岛素抵抗,其机制可能与激活SIRT1/NF-κB信号通路有关。
Objective To explore the ameliorating effect of butein on hepatic insulin resistance in diabetic rats and its possible mechanism. Methods SD rats were randomly divided into control group, diabetes group, low-dose group(Butein-L group) and high-dose group(Butein-H group) with 10 rats in each group. Fasting blood glucose and oral glucose tolerance of rats were measured. The levels of insulin in serum, total cholesterol and triglyceride, as well as the levels of TNF-α and IL-6 in liver tissue were detected by ELISA. Liver glycogen content was detected. The insulin resistance index and insulin sensitivity index of rats were calculated. The histopathological changes of liver were observed by HE staining.Western blot was used to detect the expression of IRS-1 and GLUT4 in liver tissues. The expression of SIRT1 and NF-κB in liver tissue was detected by immunofluorescence. Results Butein can reduce fasting blood glucose, serum insulin and blood lipid levels, increase liver glycogen content, reduce insulin resistance index, increase insulin sensitivity index, improve pathological damage of liver tissue, reduce the level of inflammatory factors in liver tissue, and increase the expression of IRS-1, GLUT4 and SIRT1 in liver tissue of diabetic rats, decrease the expression of NF-κB. Conclusion Butein can improve insulin resistance in diabetic rats, and the mechanism may be related to the activation of SIRT1/NF-κB signaling pathway.
