摘要
目的 探讨长链非编码RNA MALAT1对结肠癌HCT116细胞增殖和侵袭的影响并初步分析其机制。方法 体外培养结肠癌HCT116细胞,转染lncRNA MALAT1抑制物,实时定量PCR方法检测lncRNA MALAT1和miR-124的表达,双荧光素酶报告基因实验分析lncRNA MALAT1和miR-124的靶向关系,MTT法检测各组HCT116细胞增殖能力,Transwell实验检测各组HCT116细胞侵袭能力。结果 转染MALAT1抑制物能够降低HCT116细胞lncRNA MALAT1的表达,上调miR-124的表达(P<0.05),双荧光素酶实验证明miR-124是lncRNA MALAT1的靶基因。lncRNA MALAT1表达抑制后,HCT116细胞的增殖与侵袭能力下降(P<0.05)。结论 沉默lncRNA MALAT1可以抑制结肠癌细胞HCT116的增殖和侵袭,其机制可能与lncRNA MALAT1负调控miR-124表达有关。
Objective To investigate the effect and mechanism of long-chain non coding RNA MALAT1 on the proliferation and invasion of colon cancer HCT116 cells. Methods The HCT116 cells were cultured in vitro and transfected with the MALAT1 inhibitor. The level of lncRNA MALAT1 and miR-124 were detected by quantitative realtime PCR, and the target relationship between lncRNA MALAT1 and miR-124 was verified by double luciferase assay. The proliferation ability of HCT116 cells was detected by MTT. The invasion ability was detected by Transwell. Results After silencing lncRNA MALAT1, the expression of lncRNA MALAT1 was decreased and the expression of miR-124 was upregulated. Double-luciferase assay confirmed that miR-124 was the target gene of MALAT1. The inhibition of lncRNA MALAT1 expression significantly reduced the proliferation and invasion of HCT116 cells. Conclusion lncRNA MALAT1may regulate the proliferation and invasion of HCT116 cells via miR-124 pathway.
