超短波对大鼠急性肺损伤炎症反应及细胞间黏附分子-1表达的影响 被引量:2

Effects of ultrashort wave on inflammatory response and intercellular adhesion molecule-1 in rats with acute lung injury

摘要 目的观察超短波对大鼠急性肺损伤(ALI)炎症反应及细胞间黏附分子(ICAM)-1表达的影响。方法24只3月龄雄性SD大鼠随机分为对照组、ALI组、超短波组,每组8只。ALI组与超短波组大鼠通过气管内滴注脂多糖(LPS)复制急性肺损伤模型,对照组大鼠气管内滴注等量生理盐水。超短波组在大鼠滴注LPS即刻、4 h、8 h给予超短波干预,每次15 min。在气管内滴注LPS或生理盐水24 h后处死实验动物,对大鼠肺组织进行苏木素-伊红(HE)染色、肺组织损伤评分和肺组织湿/干重(W/D)比值比较;酶联免疫吸附试验(ELISA)检测血清白细胞介素(IL)-6、转化生长因子(TGF)-β水平,RT-PCR和Western印迹分别检测ICAM-1 mRNA及蛋白表达水平。结果ALI组W/D比值较对照组明显升高(P<0.01),超短波组比ALI组降低,但差异无统计学意义(P<0.05)。对照组肺组织结构正常;ALI组肺组织可见大量中性粒细胞浸润、聚集,有少量红细胞渗出,肺泡壁变厚断裂,肺泡结构破坏明显;超短波组大鼠肺泡结构相对ALI组完整,肺组织间中性粒细胞浸润明显减少,红细胞渗出减少。ALI组肺组织损伤评分较对照组明显升高(P<0.01);超短波组较ALI组明显减低(P<0.01)。ALI组IL-6较对照组明显升高(P<0.01),超短波组较ALI组明显减低(P<0.05);ALI组TGF-β较对照组明显降低(P<0.01),超短波组较ALI组明显升高(P<0.05)。ALI组ICAM-1 mRNA表达水平较对照组明显升高(P<0.01),超短波组较ALI组明显降低(P<0.01)。结论超短波可通过下调ICAM-1的表达,抑制急性肺损伤大鼠肺组织的炎症反应。 Objective To observe the effect of ultrashort wave on the inflammatory response and the expression ofintercellular adhesion molecules(ICAM)-1 in rats with acute lung injury(ALI).Methods Twenty-four 3-month-old male SD rats were randomly divided into control,ALI and ultrashort wave groups(n=8).Rats in ALI and ultrashort wave groups were given intratracheal lipopolysaccharide(LPS)to replicate ALI model.Rats in control group were given intratracheal infusion of the same amount of normal saline.The rats in ultrashort wave group were given ultrashort wave intervention immediately,4 and 8 h after LPS infusion,each time for 15 minutes.The rats were sacrificed 24 h after intratracheal infusion of LPS or normal saline.The lung tissues of rats were compared by HE staining,lung histopathological semi-quantitative score and lung wet/dry weight ratio(W/D).Serum levels of inflammatory cytokines IL-6 and TGF-βwere detected by ELISA,the expression levels of ICAM-1 protein and mRNA were detected by RT-PCR and Western-blot.Results Lung W/D in ALI group was significantly higher than that in control group(P<0.01).There was no significant difference in lung W/D between ALI group and ultrashort wave group.The lung structure of control group was normal.In ALI group,a large number of neutrophils and a small amount of red blood cells could be seen in the lung tissue;the alveolar wall thickened and fractured,and the alveolar structure was obviously damaged.The alveolar structure of ultrashort wave group was more complete than that in ALI group,the infiltration of neutrophils among lung tissues was significantly reduced,and the exudation of red blood cells was decreased.Semi-quantitative score of lung histopathology ofALI group was significantly higher than that of control group(P<0.01),that of ultrashort wave group was significantly lower than that of ALI group(P<0.01).Serum IL-6 of ALI group was significantly increased than that in control group(P<0.01),and that in ultrashort wave group was significantly decreased than that in ALI group(P<0.05).Serum TGF-βof ALI groupwas significantly lower than that of control group(P<0.01),that in ultrashort wave group was significantly higher than that of ALI group(P<0.05).The mRNA and protein expressions of ICAM-1in ALI group were significantly higher than those in control group(P<0.01),and those of ultrashort wave group were significantly lower than those in ALI group(P<0.01).Conclusions Ultrashort wave may inhibit the inflammatory response in ALI rats by down-regulating the expression of ICAM-1.
