射干提取物对AOM/DSS小鼠炎性反应相关性肠癌的干预作用及机制 被引量:2

The Effect and Mechanism of Shegan Extract on Inflammation-related Colorectal Cancer in AOM/DSS Mice

摘要 目的探讨射干提取物对AOM/DSS小鼠炎性反应相关性肠癌的干预作用和可能机制。方法以AOM/DSS制备小鼠炎性反应相关性肠癌模型,观察射干提取物对小鼠的一般情况,进行疾病活动指数(DAI)评分;检测小鼠血清中IL6和STAT3含量变化;进行结直肠段病理组织学评分,考察射干提取物对该模型的干预作用。结果造模后各组小鼠分别出现了腹泻、便血、脱肛、消瘦等表现;各DSS循环后模型组DAI评分均升高(P<0.01),第三循环后高剂量组能明显降低造模后小鼠的DAI评分(P<0.05);高剂量组肿瘤缩小;中、高剂量组小鼠腺体排列较模型组规则,炎性细胞浸润程度减轻;模型组与空白对照组比较,病理组织学评分分值明显增高(P<0.01),可见明显的炎性反应和肉芽组织;高剂量组评分值较模型组降低(P<0.05);低、中、高剂量组均可明显降低小鼠血清中IL6蛋白水平(P<0.05,P<0.01);中、高剂量组均可明显降低小鼠血清中STAT3蛋白水平(P<0.05,P<0.01)。结论高剂量组可抑制炎性反应相关性肠癌进展,其作用机制可能与抑制IL6/STAT3信号通路有关。 Objective To explore the interventional effect and possible mechanism of Shegan extract on inflammation-related bowel cancer in AOM/DSS mice.Methods Observe the general state of the mice and score the disease activity index(DAI);Detect the changes in the levels of IL6 and STAT3 in the serum;The colorectal segment was examined by histopathology and histopathology score was performed to investigate the intervention effect of Shegan extract on the model.Results After modeling,mice in each group showed diarrhea,hematochezia,rectal prolapse,weight loss,etc.;The DAI scores of the model group increased after each DSS cycle(P<0.01),and after the third cycle,the high-dose group can significantly reduce the DAI score of the mice after modeling(P<0.05);The high-dose Shegan extract group had smaller tumors;In the middle and high dose groups,the arrangement of the glands of the Shegan extract mice was more regular than that of the model group,and the degree of inflammatory cell infiltration was reduced;Compared with the control group,the histopathology scores of the model group were significantly higher(P<0.01),and obvious inflammation and granulation tissue were visible;the scores of the high-dose Shegan extract group were lower than those of the model group,and the difference was statistically significant(P<0.05);The low,medium and high doses of Shegan extract can significantly reduce the IL6 protein level in mice serum(P<0.05,P<0.01);The medium andhigh dose group can significantly reduce the STAT3 protein level in mice serum(P<0.05,P<0.01).Conclusion The high-dose Shegan extract group can inhibit the progression of inflammation-related bowel cancer,and its mechanism may be related to the inhibition of IL6/STAT3 signaling pathway.
