黄连解毒汤古法煎煮与现代煎煮的差异研究

摘要 目的:通过水煎液化学成分及转移率、出膏率、日服化学成分剂量等指标,比较黄连解毒汤古法煎煮与现代煎煮的差异,为黄连解毒汤开发研究提供参考。方法:按照古籍《外台秘要》记载的煎煮方法(“黄连三两,黄芩、黄柏各二两,栀子十四杖擘,以水六升,煮取二升”)和现代煎煮方法(“黄连9 g,黄芩6 g,黄柏6 g,栀子9 g,1200 mL水,煮取400 mL”)制备水煎液,冻干得干粉,采用高效液相色谱法,检测饮片和冻干粉中表小檗碱、黄连碱,巴马汀、小檗碱、黄柏碱、黄芩苷、栀子苷含量,计算出膏率、转移率和日服有效成分量,比较汤古法煎煮与现代煎煮的差异。结果:古法煎煮平均出膏量为17.86 g,出膏率为15.99%,现法煎煮平均出膏量为7.42,出膏率为24.34%,从出膏率看,现法煎煮优于古法煎煮,从化学指标转移率看,现法煎煮化学成分转移率均大于古法煎煮;各化学成分日服用量差异与日服生药量差异不一致,古法煎煮高于现法煎煮。结论:从化学角度出发,通过比较黄连解毒汤不同煎煮方法,为其“物质基准”研究中剂量及煎煮方法的确定提供实验参考。 Objective Through the chemical composition and transfer rate of the water decoction,the ointment rate,the daily dosage of chemical components and other indicators,the difference between the ancient decoction of Huanglian Jiedu Decoction and the modern decoction is compared,which provides a reference for the development and research of the classic prescription-Huanglian Jiedu Decoction.Methods According to the decocting method recorded in the ancient book“Wai Tai Mi Yao”(Coptis scutellaria three liang,scutellaria baicalensis,cypress each two liang,gardenia fourteen sticks,six liters of water,two liters of boiling)and modern decocting methods(Coptis chinensis 9 g,Scutellaria baicalensis 6 g,Cork 6 g,Gardenia 9 g,1200 mL water,boil 400 mL)Prepare water decoction,freeze-dried dry powder,use high performance liquid chromatography to detect epiberberine in decoction pieces and freeze-dried powder,Berberine,palmatine,berberine,phellodendron,baicalin,geniposide content,calculate the cream rate,transfer rate and daily effective ingredients,and compare the difference between ancient decoction and modern decoction.Results From a chemical point of view,by comparing different decoction methods of Huanglian Jiedu Decoction,it provides experimental reference for the determination of dosage and decoction method in its“material benchmark”study.
