健心颗粒对心力衰竭大鼠模型心肌代谢重构的作用 被引量:3

摘要 目的通过构建心力衰竭动物模型分析健心颗粒对心力衰竭心肌代谢重构的作用及机制。方法选择30只健康SD大鼠依照随机数表法分为对照组、模型组及治疗组,每组各10只。模型组及治疗组大鼠利用腹主动脉缩窄法制备心力衰竭模型,治疗组使用健心颗粒进行灌胃治疗,对照组与模型组使用等体积蒸馏水灌胃干预。大鼠清醒状态下对大鼠尾动脉收缩压(SBP)及心率进行检测,测定大鼠治疗后左心室舒张末期内径(LVEDD)、左心室收缩末期内径(LVESD)水平;给药4周后取心脏保留左心室及左心室间隔称重,以左心室重量/体重的比值计算左心室重量指数(LVMI);取大鼠左心室心肌组织,测定线粒体蛋白(OD)浓度、Na^(+)-K^(+)-ATP酶和Ca^(2+)-ATP酶活性;检测大鼠血浆中血管紧张素Ⅱ(ANGⅡ)、脑钠肽(BNP)及心钠素(ANP)水平。结果治疗组大鼠SBP明显高于模型组,心率、LVEDD及LVESD明显低于模型组,差异有统计学意义(P<0.05);治疗组大鼠体重、左心室重量及LVMI均明显高于模型组,差异有统计学意义(P<0.05);治疗组大鼠OD、Na^(+)-K^(+)-ATP酶和Ca^(2+)-ATP酶活性、ANGⅡ、BNP、ANP均明显高于模型组,差异有统计学意义(P<0.05)。结论健心颗粒可有效提升心力衰竭大鼠模型干预后心肌代谢能力,改善心肌重构,提高疗效。 Objective To analyze the effect and mechanism of Jianxin Granules on myocardial metabolic remodeling in heart failure by constructing an animal model of heart failure.Methods A total of 30 healthy SD rats were divided into the control group,the model group and the treatment group according to the random number table method,with 10 rats in each group.Rats in the model group and the treatment group were used to prepare heart failure models by abdominal aortic banding.Rats in the treatment group were gavaged with Jianxin Granules,while rats in the control group and model group were gavaged with equal volume of distilled water.The caudal artery systolic blood pressure(SBP)and heart rate of rats were measured in the awake state,and the levels of left ventricular end-diastolic diameter(LVEDD)and left ventricular end-systolic diameter(LVESD)of rats were measured aft er treatment.The l eft ventricle and l eft ventricular septum were weighed 4 weeks after administration,and the left ventricular weight index(LVMI)was calculated with the left ventricular mass/body weight ratio.The OD concentration,Na^(+)-K^(+)-ATPase and Ca^(2+)-ATPase activities were measured with the left ventricular myocardial tissue of rats.In addition,the levels of angiotensinⅡ(ANGⅡ),brain natriuretic peptide(BNP)and atrial natriuretic peptide(ANP)in the plasma of rats were measured.Results The SBP level was significantly higher and the heart rate,LVEDD and LVESD levels were significantly lower in the treatment group than those in the model group,with statistically significant differences(P<0.05).The body mass,left ventricular mass and LVMI in the treatment group were significantly higher than those in the model group,with statistically significant differences(P<0.05).The OD,Na^(+)-K^(+)-ATPase and Ca^(2+)-ATPase activities,ANGⅡ,BNP and ANP in the treatment group were significantly higher than those in the model group,with statistically significant differences(P<0.05).Conclusion Jianxin Granules can effectively strengthen myocardial metabolic capacity,improve myocardial remodeling and enhance the efficacy in rats through intervening in the heart failure model of rats.
