慢阻肺患者外周血单个核细胞lncRNA SNHG5的表达与炎症因子水平相关性研究 被引量:3

Correlation between Expression of lncRNA SNHG5 in Peripheral Blood Mononuclear Cells and Level of Inflammatory Factors in Patients with Chronic Obstructive Pulmonary Disease

摘要 目的探讨慢阻肺(COPD)患者外周血单个核细胞lncRNA SNHG5的表达与炎症因子水平相关性.方法选取COPD稳定期患者80例,急性加重期患者80例,健康志愿者80名为对照组.分离外周血单个核细胞,RT-PCR检测lncRNA SNHG5、核苷酸结合寡聚化结构域1(NOD1)、核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)的mRNA表达;Western blot检测NOD1、NLRP3的蛋白表达;酶联免疫吸附法检测血清C反应蛋白、肿瘤坏死因子α、白细胞介素-6(IL-6)表达水平.结果COPD急性加重期组lncRNA SNHG5的表达水平明显低于COPD稳定期组和对照组,COPD稳定期组明显低于对照组(P<0.05).COPD急性加重期组NOD1、NLRP3的mRNA和蛋白表达水平明显高于COPD稳定期组和对照组,COPD稳定期组明显高于对照组(P<0.05).COPD急性加重期组血清C反应蛋白、肿瘤坏死因子α、IL-6水平明显高于COPD稳定期组和对照组,COPD稳定期组明显高于对照组(P<0.05).Pearson相关性分析显示:COPD患者外周血单核细胞中lncRNA SNHG5表达与血清C反应蛋白、肿瘤坏死因子α、IL-6呈负相关关系(r=-0.719、-0.734、-0.760,均P<0.001).COPD患者外周血单核细胞中NOD1、NLRP3表达与血清C反应蛋白、肿瘤坏死因子α、IL-6呈正相关关系(r=0.736、0.750、0.728,均P<0.001;r=0.765、0.792、0.780,均P<0.001).结论慢阻肺患者外周血单核细胞中lncRNA SNHG5、NOD1、NLRP3可能通过调控炎症因子C反应蛋白、肿瘤坏死因子α、IL-6的表达参与COPD病情的发生与进展,其结果可用于临床诊断和预后评估. ObjectiveTo investigate the expression of long-chain noncoding RNA(lncRNA)SNHG5 and inflammatory regulators in peripheral blood mononuclear cells of patients with chronic obstructive pulmonary disease(COPD).Method80 patients with stable COPD,80 patients with acute exacerbation and 80 healthy volunteers were selected as the control group.Peripheral blood mononuclear cells were isolated and the mRNA expressions of lncRNA SNHG5,nucleotide binding oligomerization domain 1(NOD1)and nucleotide binding oligomerization domain like receptor protein 3(NLRP3)were detected by RT-PCR;The expression of NOD1 and NLRP3 protein was detected by Western blot;Serum C-reactive protein and tumor necrosis factor were detected by enzyme-linked immunosorbent assayα,Interleukin-6(IL-6)expression level.ResultsThe expression level of lncRNA SNHG5 in acute exacerbation group was significantly lower than that in stable COPD group and control group,and the expression level of lncRNA SNHG5 in stable COPD group was significantly lower than that in control group(P<0.05).The expression levels of NOD1,NLRP3 mRNA and protein in COPD acute exacerbation group were significantly higher than those in stable COPD group and control group,and those in stable COPD group were significantly higher than those in control group(P<0.05).The level of IL-6 was significantly higher than that of stable COPD group and control group,and that of stable COPD group was significantly higher than that of control group(P<0.05).Pearson correlation analysis showed that the expression of lncRNA SNHG5 in peripheral blood monocytes of COPD patients was correlated with serum C-reactive protein and tumor necrosis factorα,IL-6 was negatively correlated(r=-0.719,-0.734,-0.760,P<0.001).The expression of NOD1 and NLRP3 in peripheral blood monocytes of COPD patients was correlated with serum C-reactive protein and tumor necrosis factorα,IL-6 was positively correlated(r=0.736,0.750,0.728,P<0.001;r=0.765,0.792,0.780,P<0.001).ConclusionThe expression of lncRNA SNHG5 is down-regulated and the expression of NOD1 and NLRP3 are up-regulated in peripheral blood mononuclear cells of patients with COPD,which can promote the development of the disease by regulating the inflammatory response.
