基于TLR/NF-κB通路探究蓬子菜总黄酮对动脉粥样硬化模型家兔的干预效果 被引量:4

摘要 目的分析蓬子菜总黄酮对动脉粥样硬化模型家兔主动脉组织中Toll样受体(TLR)/核转录因子-κB(NF-κB)通路的影响。方法选取75只家兔,其中15只作为正常组,其余家兔建立动脉粥样硬化模型,建模成功后随机分为模型组、低剂量组、中剂量组、高剂量组,每组15只。低剂量组、中剂量组、高剂量组分别给予0.05 mg/kg、1.00 mg/kg、1.50 mg/kg蓬子菜总黄酮灌胃,模型组和正常组给予等体积生理盐水灌胃。采用酶联免疫吸附法检测白细胞介素-6(IL-6)、白细胞介素-1α(IL-1α)、肿瘤坏死因子-α(TNF-α)、三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C);采用放射免疫法检测6-酮-前列腺素(6-keto-PGF_(1α))、血栓素B_(2)(TXB_(2));采用蛋白质印迹法(Western Blot)检测TLR/NF-κB通路蛋白表达量。结果高剂量组、中剂量组、低剂量组、模型组IL-6、IL-1α、TNF-α水平均高于正常组(P<0.05);高剂量组、中剂量组、低剂量组IL-6、IL-1α、TNF-α水平低于模型组(P<0.05);高剂量组、中剂量组IL-6、IL-1α、TNF-α水平低于低剂量组(P<0.05);高剂量组IL-6、IL-1α、TNF-α水平低于中剂量组(P<0.05)。中剂量组、低剂量组、模型组6-keto-PGF_(1α)水平均低于正常组,TXB_(2)水平均高于正常组(P<0.05);高剂量组、中剂量组、低剂量组6-keto-PGF_(1α)水平高于模型组,TXB_(2)水平低于模型组(P<0.05);高剂量组、中剂量组6-keto-PGF_(1α)水平高于低剂量组,TXB_(2)水平低于低剂量组(P<0.05);高剂量组6-keto-PGF_(1α)水平高于中剂量组,TXB_(2)水平低于中剂量组(P<0.05)。中剂量组、低剂量组、模型组TG、TC、LDL-C水平均高于正常组,HDL-C水平均低于正常组(P<0.05);高剂量组、中剂量组、低剂量组TG、TC、LDL-C水平低于模型组,HDL-C水平高于模型组(P<0.05);高剂量组、中剂量组TG、TC、LDL-C水平低于低剂量组,HDL-C水平高于低剂量组(P<0.05);高剂量组TG、TC、LDL-C水平低于中剂量组,HDL-C水平高于中剂量组(P<0.05)。中剂量组、低剂量组、模型组TLR3、NF-κB p62水平高于正常组(P<0.05);中剂量组、低剂量组TLR3、NF-κB p62水平低于模型组(P<0.05);高剂量组、中剂量组TLR3、NF-κB p62水平低于低剂量组(P<0.05)。结论蓬子菜总黄酮可能通过调控TLR/NF-κB信号通路中相关重要信号传导分子的蛋白表达,发挥改善动脉粥样硬化局部炎症的作用,且表现为剂量依赖。 Objective To analyze the effect of flavone of Galium verum L(FGVL)on Toll-like receptors(TLR)/nuclear factor-κB(NF-κB)pathway in aortic tissue of rabbits with atherosclerosis.Methods Seventy-five rabbits were selected,of which 15 were used as the normal group,and the rest of the rabbits were established atherosclerosis models.After successful modeling,the rabbits were randomly divided into model group,low-dose group,medium-dose group,and high-dose group,with 15 rabbits in each group.The rabbits were given 0.05 mg/kg,1.00 mg/kg,and 1.50 mg/kg of FGVL intragastric administration,respectively.The rabbits in model group and normal group were given equal volume saline intragastric administration.The levels of interleukin-6(IL-6),interleukin-1α(IL-1α),tumor necrosis factor-α(TNF-α),triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)were detected by enzyme-linked immunosorbent assay(ELISA).Radioimmunoassay was used to detect 6-keto-prostaglandin F 1α(6-keto-PGF_(1α))and thromboxane B_(2)(TXB_(2)).The protein expression of TLR/NF-κB pathway was detected by Western Blot method.Results The levels of IL-6,IL-1α,and TNF-αin high-dose group,medium-dose group,low-dose group,and model group were higher than those in normal group(P<0.05),which in high-dose,medium-dose,and low-dose groups were lower than those in model group,with a dose-dependent manner(P<0.05).The levels of 6-keto-PGF_(1α) in medium-dose group,low-dose group,and model group were lower than that in normal group,and the level of TXB_(2) was higher than that in normal group(P<0.05).The levels of 6-keto-PGF_(1α) in high-dose,medium-dose,and low-dose groups were higher than that in model group,while the levels of TXB_(2) were lower than that in model group,with a dose-dependent manner(P<0.05).The levels of TG,TC,and LDL-C in medium-dose group,low-dose group,and model group were higher than those in normal group,while the HDL-C levels were lower than that in normal group(P<0.05).The levels of TG,TC,and LDL-C in high-dose,medium-dose,and low-dose groups were lower than those in model group,while the HDL-C levels were higher than that in model group,and with a dose-dependent manner(P<0.05).The levels of TLR3 and NF-κB p62 in medium-dose group,low-dose group,and mode l group were higher than those in normal group(P<0.05),which in high-dose,medium-dose,and low-dose groups were lower than those in model group(P<0.05),and which in high-dose and medium-dose groups were lower than those in low-dose group(P<0.05).Conclusion FGVL may play a role in improving local inflammation of atherosclerosis,with a dose dependent manner by regulating the protein expression of important signal transduction molecules in TLR/NF-κB signaling pathway.
