摘要
目的:研究腹部推拿对非酒精性脂肪性肝病(NAFLD)大鼠肠道黏膜的蛋白闭锁连接蛋白-1(ZO-1)、E-钙黏蛋白(E-Cadherin)、多聚体纤维状肌动蛋白(F-actin)蛋白及超微结构的影响。方法:建立NAFLD大鼠模型,随机分为腹部推拿组和模型对照组,每组9只,另设9只大鼠为正常对照组。腹部推拿组每天推拿干预10 min;模型对照组每日束缚大鼠,10 min后解开束缚;正常对照组不予任何干预手段。干预4周后,利用免疫组化方法检测肠黏膜F-actin、ZO-1、E-Cadherin蛋白表达;同时,利用透射电镜观察大鼠的肠黏膜组织超微结构。结果:造模后F-actin和ZO-1蛋白免疫组化染色为阴性;腹部推拿干预后,F-actin和ZO-1蛋白免疫组化染色为阳性;而E-Cadherin蛋白在造模后及腹部推拿干预后,染色结果均为阳性;同时,通过透射电镜观察肠道黏膜组织,造模后细胞结构及线粒体等细胞器超微结构被破坏,而腹部推拿组通过干预可有一定程度的结构改善。结论:腹部推拿可上调F-actin和ZO-1蛋白含量,修复肠道黏膜组织结构,以改善NAFLD大鼠肠道黏膜通透性,逆转NAFLD的脂肪变性。
Objective:To study the effects of abdominal tuina on the expression of ZO-1,E-cadherin,F-actin,and the ultrastructural in intestinal mucosa of rats with non-alcoholic fatty liver disease(NAFLD).Methods:NAFLD rat models were induced by high fat diet.The rats were divided into abdominal Tuina group and model group by random number table,with 9 rats in each group.Another 9 rats were set as normal control group.The abdominal tuina group was given tuina intervention every day for 10 minutes,while the model group was confined to the experimental platform every day and took supine position without any intervention,and the rats were released after 10 minutes.The normal control group did not receive any intervention.After 4 weeks of continuous intervention,the expressions of F-actin,ZO-1 and E-cadherin in intestinal mucosa were detected by immunohistochemistry;The ultrastructure of intestinal mucosa in rats was observed by transmission electron microscope.Results:After modeling,F-actin and ZO-1 protein were negative by immunohistochemical staining;After abdominal tuina intervention,F-actin and ZO-1 protein immunohistochemical staining was positive;The staining results of E-cadherin protein were positive after modeling and after abdominal massage intervention;The intestinal mucosa was observed by transmission electron microscope.After modeling,the cell structure and ultrastructure of mitochondria and other organelles were destroyed,while the abdominal tuina group had a certain degree of structural improvement through intervention.Conclusion:Abdominal tuina can up regulate the'contents of F-actin and ZO-1 protein,repair the structure of intestinal mucosa,improve the intestinal mucosal permeability and reverse the fatty degeneration of NAFLD rats.
