摘要
目的观察麦粒灸对甲状腺功能减退(简称甲减)大鼠免疫应答的影响,探究麦粒灸对甲状腺功能减退大鼠的保护作用。方法将30只SD大鼠按照随机数字表法分为空白组、模型组、麦粒灸组,共3组。除空白组外,其余组用丙硫氧嘧啶灌胃的方法进行甲减疾病造模。麦粒灸组采用麦粒柱于大椎、脾俞(双)、肾俞(双)、命门施灸,持续4周。治疗结束后,取脾脏,后分离胸腺和甲状腺,取血清用ELISA方法检测游离三碘甲状腺原氨酸(free triiodothyronine,FT3)、游离甲状腺素(free thyroxine,FT4)、促甲状腺素(thyrotropin,TSH)、甲状腺过氧化物酶抗体(thyroid peroxidase antibody,TPOAb)、甲状腺球蛋白抗体(anti-thyroglobulin antibodies,TGAb)、干扰素(interferonγ,I IFN-γ)、白介素10(interleukin 10,IL-10)、转化生长因子β(transforming growth factor-β,TGF-β);并运用免疫组化染色酶法检验组织中的细胞炎性因子(IL-10、IFN-γ、TGF-β)水平。结果空白组、模型组和麦粒灸组大鼠干预后体质量分别为(322.93±37.13)g、(227.30±30.26)g、(284.60±32.25)g。与模型组相比,麦粒灸组大鼠血清中游离甲状腺素(FT4)浓度升高(P<0.05),TSH浓度下降(P<0.05);同时,麦粒灸组大鼠TGAb、IFN-γ及TGF-β浓度下降(P<0.05),IL-10浓度升高(P<0.05)。在组织中,麦粒灸组IFN-γ相较模型组明显下降(P<0.05)。结论甲状腺功能减退症存在着免疫应答失调的状况,麦粒灸可能通过良性调控TGAb滴度,干预促炎因子IFN-γ和TGF-β的表达,增强抗炎因子IL-10的表达来缓解机体免疫紊乱状况,从而改善甲状腺功能减退。
Objective To observe the effect of wheat grain moxibustion on immune response of hypothyroidism rats,and to explore the protective effect of wheat grain moxibustion on hypothyroidism rats.Methods 30 SD rats were randomly divided into the blank group,the model group and the wheat grain moxibustion group,ten in each group.The hypothyroidism rat model was established by intragastric administration of PTU in the model group and the wheat grain moxibustion group.The wheat grain moxibustion group was treated with wheat grain moxibustion on Dazhui,Pishu(double),Shenshu(double)and Mingmen for 4 weeks.After the treatment,the spleen,thymus and thyroid gland were separated,and the serum levels of free triiodothyronine(FT3),free thyroxine(FT4),thyrotropin(TSH),thyroid peroxidase antibody(TPOAb),thyroglobulin antibody(TGAb),interferon(IFN-γ),interleukin 10(IL-10)and transforming growth factor-β(TGF-β)were detected by ELISA.The cellular inflammatory factors(IL-10,IFN-γ,TGF-β)in the tissues were detected by immunohistochemical(IHC)staining enzyme method.Results The body weights of the blank group,the model group and the wheat moxibustion group were(322.93±37.13),(227.30±30.26)and(284.60±32.25)g.After the intervention of wheat moxibustion,the contents of free thyroxine(FT4)were increased(P<0.05),while the contents of thyroid stimulating hormone(TSH)were decreased(P<0.05).The expression of serum thyroglobulin antibody(TGAb),interferon-γ(IFN-γ)and transforming growth factor-β(TGF-β)were decreased(P<0.05),while the expression of IL-10 were increased(P<0.05).The level of IFN-γin thyroid tissues were decreased(P<0.05).Conclusion Dysimmune response exists in hypothyroidism.Wheat grain moxibustion may relieve the immune disorder by benign regulation of TG-Ab titer,interfering with the expression of Pro-inflammatory factors IFN-γand TGF-β,and enhancing the expression of anti-inflammatory factor IL-10,thus improving hypothyroidism.
