摘要
血脑屏障通透性主要与脑血管内皮细胞间的紧密连接、控制物质外排的P糖蛋白(P-glycoprotein,P-gp)转运体、内皮细胞间隙的孔穴以及胞吞、胞饮作用有关。芳香开窍中药由于自身脂溶性成分较易透过血脑屏障;麝香、冰片、石菖蒲能够抑制P-gp的外排作用,提升药物进入脑内浓度,亦可抑制Claudin-5蛋白表达,使血管内皮细胞紧密连接的缝隙变宽等,增加血脑屏障通透性;石菖蒲、冰片亦可增加五羟色胺(5-hydroxytryptamine,5-HT)含量,提高受体结合率,促进内皮细胞收缩致紧密连接打开,增加血脑屏障通透性;苏合香、安息香通过影响P-gp的外排作用,增加血脑屏障通透性。在病理状态时,麝香、冰片、苏合香、安息香通过降低炎症因子的含量,减缓该因子对内皮细胞间的紧密连接破损,麝香亦可抑制基质金属蛋白酶的表达、减缓自由基引起的细胞损伤从而稳定血脑屏障的结构,以降低血脑屏障通透性;石菖蒲通过发挥抗自由基损伤作用降低血脑屏障通透性。本文对芳香开窍中药对血脑屏障通透性调控作用进行研究。为今后研究单用或联合此类药物治疗脑部疾病提供理论基础。
The permeability of blood-brain barrier is mainly related to the tight junction between cerebral vascular endothelial cells,P-glycoprotein(P-gp)transporters that control the efflux of substances,holes in the intercellular space,endocytosis and endocytosis.Because the fat-soluble components of aromatic Kaiqiao herbs can easily pass through the blood-brain barrier,musk,borneol and Acorus calamus can inhibit the efflux of P-gp,increase the concentration of drugs into the brain,inhibit the expression of Claudin-5 protein,widen the gap of tight junction of vascular endothelial cells,and increase the permeability of blood-brain barrier.Acorus tatarinowii and borneol can also increase the content of 5-HT and the rate of receptor binding,promote the opening of tight junction caused by endothelial cell contraction,and increase the permeability of blood-brain barrier.Both Storesin and benzoin can increase the permeability of blood-brain barrier by affecting the efflux of P-gp.In the pathological state,musk,borneol,Storesin and benzoin slow down the damage of tight junction between endothelial cells by reducing the content of inflammatory factors.Musk can also inhibit the expression of matrix metalloproteinases and slow down the cell damage caused by free radicals so as to stabilize the structure of blood-brain barrier and reduce the permeability of blood-brain barrier.Acorus calamus can reduce the permeability of blood-brain barrier by anti-free radical injury.In this paper,the regulatory effect of aromatic Kaiqiao herbs on the permeability of blood-brain barrier was studied.The purpose of this study is to provide a theoretical basis for the future study of single or combined use of these drugs in the treatment of brain diseases.
