摘要
试验旨在探讨适合菲牛蛭中小分子多肽的Tricine-SDS-PAGE分离方法。采用电泳纯重组水蛭素验证Tricine-SDS-PAGE的分离效果,对菲牛蛭的水提醇沉粗提物进行分离,建立小分子多肽对数分子量和迁移率的回归关系方程,计算多肽分子质量。结果显示:菲牛蛭中小分子多肽对数分子量和迁移率成正比直线关系,从菲牛蛭中分离得到7个分子量在10~31 kDa的多肽。研究表明,Tricine-SDS-PAGE对菲牛蛭中的小分子多肽具有较好的分离效果。
The experiment was to explore Tricine-SDS-PAGE separation method suitable for small molecule polypeptides in Hirudinari manillensis.The separation effect of Tricine-SDS-PAGE was verified by electrophoresispurified recombinant hirudin,and the crude extract of water-extracted and Hirudinari manillensis was separated,and the regression equation of small molecular polypeptide logarithmic molecular weight and mobility was established,calculate the molecular mass of the peptide.The results showed that there was a linear relationship between the logarithmic molecular weight and the mobility of small-molecule polypeptides in Hirudinari manillensis.Seven peptides with molecular weights ranging from 10~31 k Da were isolated from Hirudinari manillensis.The study indicates that TricineSDS-PAGE has a good separation effect on small molecular polypeptides in leech.
