摘要
目的:建立同时测定壮药三角泡槲皮素、木犀草素、芹菜素3种黄酮成分含量的方法,优化三角泡黄酮的提取工艺,并对其抗氧化活性进行研究。方法:以乙醇浓度、料液比、提取时间及提取温度为考察因素,以3种黄酮提取值的总量为考察指标,采用响应面法优选最佳的超声提取工艺,测定三角泡提取物清除DPPH自由基、ABTS^(+)自由基能力及羟基自由基的能力,研究其抗氧化作用。结果:确定最优提取条件为80.00%,料液比为1:37.49,提取时间为41.18 min,提取温度为68.36℃,在此条件下三角泡中槲皮素、木犀草素和芹菜素的总提取值为4.315 mg/g。三角泡提取物清除DPPH自由基、ABTS^(+)自由基及羟基自由基的IC_(50)值分别为46.99、45.72、44.21 mg/mL。结论:本试验建立的HPLC法适用于三角泡中3种黄酮类成分的同时测定,响应面法优化的三角泡黄酮提取工艺准确可靠,三角泡具有一定的体外抗氧化活性。
Objective:To establish a method for the determination of the contents of quercetin,apigenin and luteolin,and optimize the extraction process of flavonoid compounds in Sanjiaopao(Cardiosperminum halicacabum Linn),and study its anti-oxidant activity.Methods:Taking ethanol concentration,solid-liquid ratio,extraction time and extraction temperature as the investigation factors,and taking the total amount of the extraction values of three flavonoids as the investigation index,the response surface methodology was used to optimize the optimal ultrasonic extraction process,and the DPPH free radical scavenging,ABTS^(+) free radical scavenging and hydroxyl free radical scavenging abilities of the extract of Sanjiaopao(Cardiosperminum halicacabum Linn)were determined,and study its antioxidant effect.Results:The optimal extraction conditions were determined as follows:ethanol concentration was 80.00%,solid-liquid ratio was 1∶37.49,extraction time was 41.18 min,and extraction temperature was 68.36℃.The total extraction value was 4.315 mg/g.Under these conditions,the total extraction value of quercetin,luteolin and apigenin in Sanjiaopao(Cardiosperminum halicacabum Linn)was 4.315 mg/g.The IC_(50) values of DPPH free radical,ABTS^(+) free radical and hydroxyl free radical scavenging by the extract of Sanjiaopao(Cardiosperminum halicacabum Linn)were 46.99,45.72 and 44.21 mg/mL,respectively.Conclusion:The HPLC method established in this study was suitable for the simultaneous determination of three flavonoids in Sanjiaopao(Cardiosperminum halicacabum Linn).The extraction process of flavonoids from Sanjiaopao(Cardiosperminum halicacabum Linn)optimized by response surface methodology was accurate and reliable,and Sanjiaopao(Cardiosperminum halicacabum Linn)had certain in vitro antioxidant activity.
