摘要
试验旨在考察离子液体种类对甘蔗叶总黄酮得率的影响。采用超声辅助离子液体提取法提取甘蔗叶总黄酮,以单因素试验为基础,采用正交试验设计优化提取工艺。结果显示:三丁基乙基溴化膦可以作为提取甘蔗叶总黄酮优选离子液体,经正交试验优化的最佳工艺参数为离子液体浓度0.8 mol/L、液料比25 mL/g、超声时间45 min、超声温度60℃,该条件下甘蔗叶总黄酮得率为3.53%。采用正丁醇对甘蔗叶黄酮进行萃取纯化,萃取纯化的甘蔗叶黄酮总含量达40.50%。研究表明,离子液体提取法可以高效提取甘蔗叶总黄酮,正丁醇作为萃取剂可对甘蔗叶总黄酮进行初步纯化。
The experiment was to investigate the effect of different ionic liquids on the yield of total flavonoids from sugarcane leaves. The total flavonoids from sugarcane leaves were extracted by ultrasonic-assisted ionic liquid extraction.Based on single factor experiment, the extraction process was optimized by orthogonal experimental design. The results showed that the optimal technological parameters were concentration of ionic liquid 0.8 mol/L, liquid-solid ratio 25 mL/g,ultrasonic time 45 min and ultrasonic temperature 60 ℃. Under these conditions, the yield of total flavonoids from sugarcane leaves was 3.53%. Flavonoids from sugarcane leaves were extracted and purified with n-butanol, and the total content of flavonoids from sugarcane leaves reached 40.50%. The study indicates that ionic liquid extraction method can effectively extract total flavonoids from sugarcane leaves, and n-butanol can be used as extraction agent to preliminarily purify total flavonoids from sugarcane leaves.
