Optimization of Tissue Culture and Rapid Propagation of Rhododendron hainanense

摘要 以海南杜鹃(Rhododendron hainanense)的种子为外植体,通过探究不同植物生长调节剂及其浓度对海南杜鹃不同生长阶段的影响,优化海南杜鹃的组织快繁体系,得到海南杜鹃各生长阶段组织培养的最佳培养基配方。结果表明,海南杜鹃最佳培养基pH值均为5.8,其中种子萌发最佳培养基为WPM+2.0 mg/L GA3;最佳增殖培养基为WPM+1.0 mg/L IBA+0.2 mg/L TDZ,不定芽诱导率达100%,平均增殖倍数达5.05;最佳继代培养基为WPM+0.4 mg/LIBA+0.2 mg/LTDZ+1%AC (碳源),一个月生长量最高达0.96 cm,并且植株健康;最佳生根培养基为WPM+1.0 mg/L IBA+1.5 mg/L NAA,生根率达94.44%,平均生根数量达23.48个;移栽基质为在腐殖土:椰糠=1:1 (V:V)的基础上铺一层1 cm的水苔,移栽成活率达到75.0%。本研究为杜鹃工厂化育苗和种质资源保育提供参考依据。 Tissue culture and regeneration of Rhododendron hainanense were established using the seeds as explants.By exploring the effects of different plant growth regulators and their concentrations on different growth stages of Hainan Rhododendron,the tissue rapid propagation system of Hainan Rhododendron was optimized,and the best medium formula for tissue culture of Hainan Rhododendron at each growth stage was obtained.The results shows that the optimal medium pH value was 5.8,and the optimal proliferation medium was WPM+1.0 mg/L IBA+0.2 mg/L TDZ,ionduction percentage of adventitious bud was up to 100%,and the average proliferation multiple was up to5.05;The optimal subculture medium was WPM+0.4 mg/L IBA+0.2 mg/L TDZ+1% AC,the seedlings growth status were best and the average elongation reached 0.96 cm;The best rooting medium was WPM+1.0 mg/L IBA+1.5 mg/L NAA,the rooting rate was up to 94.44%,the average rooting number was up to 23.48.Transplant medium was 1 cm sphagna based on humus soil:coco coir=2:1(V:V),and the survival rate was up to 75.0%.The study provide a reference basis for plant seedling cultivation and germplasm conservation of Rhododendron hainanense.
